枣实蝇( Carpomya vesuviana Costa)是一种新入侵检疫性有害生物。应用PCR-RFLP技术,快速区分鉴定了枣实蝇与我国口岸截获频率较高的5种检疫性实蝇。研究表明:设计出的2对引物对6种供试实蝇线粒体DNA ( mtDNA)进行PCR扩增,其扩增片断大小分别为350 bp和450 bp。 PCR扩增产物用2种限制性内切酶Dra I和MseI进行酶切,根据不同的酶切位点准确区分了6种供试的实蝇。此方法可用于口岸截获实蝇的快速检疫鉴定。

Carpomya vesuviana Costa has been a new quarantine pest in China. In this paper, the method of rapid identi-fication of C. vesuviana Costa and 5 relative species of fruit flies was studied based on PCR-RFLP of the mitochondrial DNA( mtDNA) . The results showed that DNAs, which were estimated about 350 bp and 450 bp respectively, were ob-tained by means of PCR with two pairs of primers designed from mtDNA of target pests.The PCR-products could be reg-istered by restriction enzymes ( Dra I and Mse I) . All the 6 species could be discriminated according to the banding pat-terns of the restriction enzymes. It is suggested that the method is reliable for quick discrimination of fruit flies.
