干旱是植物生长和发育的限制因素之一,因此抗旱基因克隆和功能研究具有重要的理论意义和应用价值。文章利用甘露醇模拟干旱胁迫,以发芽为筛选指标,从拟南芥突变体库中筛选鉴定了抗旱突变体csm1-1。生理生化分析表明,csm1-1抗旱性与其电导率、可溶性糖质量比和脯氨酸积累有关。利用TAIL-PCR技术和RT-PCR分析发现,CSM1基因(RCI3)编码一个过氧化物酶。研究结果为进一步研究该基因调控植物抗旱的机理提供了基础。

Drought is one of the limiting factors for plant grow th and development ,and therefore the cloning of novel drought-tolerant genes is very necessary . The wild-type and mutant lines of Arabidopsis were treated with mannitol to simulate drought stress to screen for drought-resistant mu-tants by germination analysis .One drought-tolerant csm1-1 mutant was isolated .The results of physi-ological and biochemical analysis showed that the enhanced drought tolerance of csm1-1 was related with lower conductivity and increased accumulation of soluble sugars and proline .By using TAIL-PCR technology and RT-PCR analysis ,it was found that CSM1 was the RCI3 gene encoding a peroxidase . These results provide a basis for further research on the mechanism of plant drought stress responses .
