为了研究马尾松(Pinus massoniana Lamb.)4CL 基因的遗传进化及系统发育情况,根据NCBI 上已登录的4CL 基因的序列,比对分析,利用同源序列设计引物,以马尾松基因组 DNA 为模板,扩增核酸序列,与其他21种松属植物4CL 基因进行 SNP 对比分析。结果表明:扩增出474 bp 核酸序列,松属植物4CL 基因共存在28个 SNP 位点,总核苷酸多样性θw=0.03483、πT=0.03621,具较高的多态性。系统进化树聚类结果表明,22个松属植物均集中在第一、二大类,马尾松、火炬松、班克松和小干松聚为第一大类中的第二亚类,亲缘关系较近。
為瞭研究馬尾鬆(Pinus massoniana Lamb.)4CL 基因的遺傳進化及繫統髮育情況,根據NCBI 上已登錄的4CL 基因的序列,比對分析,利用同源序列設計引物,以馬尾鬆基因組 DNA 為模闆,擴增覈痠序列,與其他21種鬆屬植物4CL 基因進行 SNP 對比分析。結果錶明:擴增齣474 bp 覈痠序列,鬆屬植物4CL 基因共存在28箇 SNP 位點,總覈苷痠多樣性θw=0.03483、πT=0.03621,具較高的多態性。繫統進化樹聚類結果錶明,22箇鬆屬植物均集中在第一、二大類,馬尾鬆、火炬鬆、班剋鬆和小榦鬆聚為第一大類中的第二亞類,親緣關繫較近。

To explore the genetic evolution and phylogenetic of 4CL gene family from P .massoniana, the 4CL gene in P .massoniana was cloned,during which,the consensus sequences of previous publish in NCBI were used to design the primers and PCR was performed on genomic DNA of P .massoniana.The results showed that there were 28 single nucleotide polymorphisms (SNP)marker between the 22 4CL gene sequences of genus Pinus.The average nucleotide diversity for the sequenced regions was calculated to beθw=0.034 83 and πT =0.036 21,with higher polymorphism.Alignment of sequences of 4CL gene with MEGA 5.2 indicated that 22 genus Pinus were clustered together to two major groups,showed a low genetic divergence among the populations of 22 genus Pinus in their evolution process,but there was a significant interspecific difference. The closer relationship was for P . massoniana, P . taeda, P .banksiana and P .contorta.The evolution of 4CL gene in the process of plant evolution were relatively conservative,because of extensive distribution of pine,so polymorphism of 4CL gene was high,the divergence should be fully considered in the 4CL gene research.
