为了研究阿勒泰大尾羊和小尾寒羊不同脂肪组织及其他组织之间基因表达的差异,探讨脂肪与肥胖相关基因(fat mass and obesity associated gene,FTO)与绵羊体脂沉积的相关性。随机各选择6只6月龄雄性阿勒泰大尾羊和小尾寒羊,禁食24 h 后屠宰,分别采集下丘脑、海马、背最长肌、心肌和脂肪组织样本,应用冰冻组织切片技术,测定脂肪细胞的面积,用 real-time PCR 检测 FTO 基因在各组织中的表达水平。结果显示,阿勒泰大尾羊尾脂脂肪细胞面积极显著高于肾周脂脂肪细胞面积(P <0.01)。阿勒泰大尾羊与小尾寒羊下丘脑和海马 FTO 基因表达水平无显著差异,背最长肌、心肌 FTO 基因表达水平阿勒泰大尾羊的极显著高于小尾寒羊的(P <0.01),肾周脂中 FTO 基因表达水平阿勒泰大尾羊的显著高于小尾寒羊的(P <0.05)。阿勒泰大尾羊肾周脂、心周脂和尾脂中 FTO 基因表达水平在肾周脂、心周脂、尾脂之间无显著差异。结果表明,FTO 基因表达水平在阿勒泰大尾羊和小尾寒羊之间存在品种间的差异。

위료연구아륵태대미양화소미한양불동지방조직급기타조직지간기인표체적차이,탐토지방여비반상관기인(fat mass and obesity associated gene,FTO)여면양체지침적적상관성。수궤각선택6지6월령웅성아륵태대미양화소미한양,금식24 h 후도재,분별채집하구뇌、해마、배최장기、심기화지방조직양본,응용빙동조직절편기술,측정지방세포적면적,용 real-time PCR 검측 FTO 기인재각조직중적표체수평。결과현시,아륵태대미양미지지방세포면적겁현저고우신주지지방세포면적(P <0.01)。아륵태대미양여소미한양하구뇌화해마 FTO 기인표체수평무현저차이,배최장기、심기 FTO 기인표체수평아륵태대미양적겁현저고우소미한양적(P <0.01),신주지중 FTO 기인표체수평아륵태대미양적현저고우소미한양적(P <0.05)。아륵태대미양신주지、심주지화미지중 FTO 기인표체수평재신주지、심주지、미지지간무현저차이。결과표명,FTO 기인표체수평재아륵태대미양화소미한양지간존재품충간적차이。
This present study was conducted to determine the difference of fat mass and obesity associated (FTO)gene expression in Aletai sheep and little-tailed Han sheep.Twelve 6-month-old male Aletai sheep and little-tailed Han sheep were respectively slaughtered.The hypothalamus,hippocampus,longissimus dorsi muscle,cardiac muscle and adipose tissue were sampled,the adipocyte area and FTO gene expression were observed through using cryo-sectioning technique and real-time PCR.The results showed that adipo-cyte area of tail fat was significantly bigger than that of perirenal fat (P <0.01).Expression of FTO gene in hypothalamus and hippocampus were no significant differences between Aletai sheep and little-tailed Han sheep.However,FTO gene in longissimus dorsi muscle (P <0.01)and cardiac muscle (P <0.01), as well as perirenal fat (P <0.05)was significantly higher in Aletai sheep compared to little-tailed Han sheep.These results suggest that expression of FTO gene had significant differences between Aletai sheep and little-tailed Han sheep.
