目的:研究一种便捷、高效地检测黄瓜花叶病毒(CMV)的方法。方法:利用表面等离子体共振(SPR)技术检测CMV。首先用11-MUA修饰SPR金片,再用EDC/NHS活化,之后通过NHS酯基与CMV抗体结合,用BSA封闭未结合的NHS酯基。将SPR金片装入SPR仪,通入待检样品,通过折射率变化实时监测实验过程。结果:该方法检测CMV的灵敏度能够达到10 ng/mL,具有良好的特异性,与同属的花生矮化病毒、番茄不孕病毒无交叉反应。结论:建立的SPR方法操作简单、灵敏度高、特异性好,是一种新的高效检测CMV的方法。

목적:연구일충편첩、고효지검측황과화협병독(CMV)적방법。방법:이용표면등리자체공진(SPR)기술검측CMV。수선용11-MUA수식SPR금편,재용EDC/NHS활화,지후통과NHS지기여CMV항체결합,용BSA봉폐미결합적NHS지기。장SPR금편장입SPR의,통입대검양품,통과절사솔변화실시감측실험과정。결과:해방법검측CMV적령민도능구체도10 ng/mL,구유량호적특이성,여동속적화생왜화병독、번가불잉병독무교차반응。결론:건립적SPR방법조작간단、령민도고、특이성호,시일충신적고효검측CMV적방법。
Objective: To research into a convenient and efficient detection methods of cucumber mosaic virus (CMV). Methods: Detection of CMV using surface plasmon resonance(SPR). First, we used 11-MUA to modify SPR gold crystal, then used EDC/NHS to active 11-MUA, followed by antibody binding to NHS esters. The un-bound NHS esters were blocked with BSA. Put SPR gold crystal into the SPR instrument, then inject into sample to be tested, we can monitor the experiment in real-time by the refractive index changes. Results: The detection limit of this method was 10 ng/mL. Also it had good specificity which had no cross reaction with peanut stunt vi-rus and tomato aspermy virus. Conclusion: The experimental results showed that this SPR method was simple op-eration and had high sensitivity and good specificity. We established a new efficient method for detection of CMV.
