目的:实验探讨Lipostabil药物主要成分对脂肪溶解效果及安全性研究。方法用5%磷脂酰胆碱(PC)、4.75%脱氧胆酸盐(DC)及Lipostabil(Li),分别作用于脂肪细胞,采用噻唑蓝(MTT)法测不同时间脂肪细胞增殖活性,酶法检测培养液三酸甘油酯(TC)含量评价脂肪细胞溶解程度。将12只Hartley纯白豚鼠,随机分为PC、DC、Li及对照组4组,分别注射不同溶脂剂和生理盐水0.5 ml于豚鼠不同部位的脂肪层中,于不同时间点切取组织作病理分析。结果三种溶脂剂对脂肪细胞增殖活性有显著降解作用,与对照组比较P<0.05,对脂肪细胞溶解作用明显,TC含量分别为PC组(4.18±0.94)mmol/L、DC组(3.92±0.74)mmol/L、Lipostabil组(4.26±0.82)mmol/L,显著高于对照组(1.92±0.14)mmol/L (P<0.05);豚鼠体内实验表明三种溶脂剂对脂肪组织有明显溶解作用,注射部位均有不同程度的肿胀、淋巴细胞浸润、脂肪细胞变性融合减少。结论 Lipostabil主要成分对脂肪细胞溶解均有一定的作用,联合优于单种溶脂剂,均对脂肪组织不具有特异性,临床局部注射溶脂应持谨慎态度。

목적:실험탐토Lipostabil약물주요성분대지방용해효과급안전성연구。방법용5%린지선담감(PC)、4.75%탈양담산염(DC)급Lipostabil(Li),분별작용우지방세포,채용새서람(MTT)법측불동시간지방세포증식활성,매법검측배양액삼산감유지(TC)함량평개지방세포용해정도。장12지Hartley순백돈서,수궤분위PC、DC、Li급대조조4조,분별주사불동용지제화생리염수0.5 ml우돈서불동부위적지방층중,우불동시간점절취조직작병리분석。결과삼충용지제대지방세포증식활성유현저강해작용,여대조조비교P<0.05,대지방세포용해작용명현,TC함량분별위PC조(4.18±0.94)mmol/L、DC조(3.92±0.74)mmol/L、Lipostabil조(4.26±0.82)mmol/L,현저고우대조조(1.92±0.14)mmol/L (P<0.05);돈서체내실험표명삼충용지제대지방조직유명현용해작용,주사부위균유불동정도적종창、림파세포침윤、지방세포변성융합감소。결론 Lipostabil주요성분대지방세포용해균유일정적작용,연합우우단충용지제,균대지방조직불구유특이성,림상국부주사용지응지근신태도。
Objective To evaluate the efficacy and safety of the main components of Lipostabil used for fat-dissolving. Methods Adipocytes were treated respectively with 5% of phosphatidylcholine (PC), 4.75%deoxycholate (DC) and Lipostabil (Li).The proliferative activity of adipocytes was evaluated by MTT at different time. The extent of adipocytes dissolution was evaluated by enzymatic measurements of triglycerides(TC). 12 Hartley guinea pigs were randomly divided into 4 groups:PC group, DC group, Li group and Comparison group, injected with 0.5 ml 5% phosphatidylcholine, 0.5 ml 4.75% deoxycholate, 0.5 ml Lipostabil and 0.5 ml normal saline into different parts of fat layer. Cut tissues at different time for histological analysis. Results Compared with control group, all these three fat dissolving agents had a notable effect on the proliferative activity of adipocytes(P<0.05). The level of TC in these three groups, PC(4.18±0.94)mmol/L, DC(3.92±0.74)mmol/L, Lipostabil(4.26± 0.82)mmol/L, were much higher than Comparison group(1.92±0.14)mmol/L(P<0.05). In vivo experiments showed a reduce in tissue swelling, lymphocytic infiltration, degeneration and incorporation of adipocytes. Conclusion All components of Lipostabil play a role in adipocyte dissolution to some extent but the effect of the combination of all components is more notable than each of them. The main components of lipostabil can also have an influence on other tissues, not only adipose tissue, which alerts clinicians to pay much more attention to injection.
