目的:探讨p130 cas在非小细胞肺癌中的表达与E-cadherin/β-catenin复合体的关系及对肿瘤细胞侵袭能力的影响。方法应用免疫组化方法检测p130 cas在145例非小细胞肺癌配对癌旁正常肺组织中的表达,并分析其表达与E-cadherin及β-catenin表达的相关性;同时进一步通过siRNA技术在肺癌细胞系中下调p130 cas的表达,观察E-cadherin和β-catenin表达的变化及肺癌细胞侵袭能力的变化。结果 p130 cas在非小细胞肺癌组织标本中表达上调(P<0.001),并与E-cadherin和β-catenin的表达下调相关(P=0.001和P=0.003);Western blot结果显示在肺癌细胞系A549和H1299中下调p130 cas的表达可以使E-cadherin和β-catenin的表达上调,而且侵袭实验结果显示下调p130 cas的表达可以使肺癌细胞的侵袭能力下降。结论 p130 cas在肺癌中可能通过下调E-cadherin/β-catenin复合体的表达促进肿瘤细胞的侵袭和转移。

Objective To investigate the relationships between p130 cas expression and E-cadherin/β-catenin adhesive complex and to explore their effects on the invasiveness of NSCLC. Methods Immunohistochemistry was used to examine the expression of p130 cas, E-cadherin andβ-catenin in 145 lung cancer tissues and paired adjacent normal lung tissues. p130 cas specific siRNA was introduced into lung cancer cell lines, western blot and transwell were used to evaluate the changes of E-cadherin and β-catenin expression and cancer cells’ invasive capabilities. Results p130 cas expression was upregulated in NSCLC tissue samples (P<0.001). The p130 cas expression was correlated with loss of E-cadherin andβ-catenin expression (P=0.001 and P=0.003, respectively). Western blotting results showed that downregulated p130 cas expression by siRNA transfection could restore both E-cadherin andβ-catenin expression and transwell test results showed that inhibit the invasive capabilities of lung cancer cells. Conclusion These results indicate that p130 cas may facilitate invasiveness and metastasis of tumor cell through its repression on E-cadherin andβ-catenin expression in NSCLC.
