目的:通过研究痛风患者载脂蛋白C-Ⅲ(ApoC-Ⅲ)启动子的单核苷酸多态性与高甘油三酯(HTG)血症之间的关系,探讨该基因变异与痛风病程进展的关系。方法测定中国山东地区300例痛风患者及300例健康人群中的ApoC-Ⅲ启动子的两个位点C-482T和T-455C的基因型和等位基因频率,同时测定两组人群的血脂水平,并对其进行比较。结果痛风组较对照组TG水平增高,差异有统计学意义(t=6.75,P<0.01)。455位点的CC、CT和TT基因型在痛风组和对照组中分布频率分别为23.0%、24.0%、53.0%和30.3%、13.3%、56.3%,差异有统计学意义(χ2=12.47,P<0.01);455位点等位基因 C 的分布频率在痛风组和对照组中49.5%和58.5%,差异有显著性统计学意义(χ2=9.78,P<0.01);482位点的CC、CT和TT基因型在痛风组和对照组中分布频率分别为28.0%、23.0%、49.0%和16.0%、33.7%、50.3%,差异有显著性统计学意义(χ2=15.89,P<0.01);482位点等位基因T的分布频率在痛风组和对照组中47.5%和58.8%,差异有统计学意义(χ2=15.47,P<0.01)。结论 ApoC-Ⅲ启动子的455和482位点的基因多态性与痛风患者的HTG发病有关。

목적:통과연구통풍환자재지단백C-Ⅲ(ApoC-Ⅲ)계동자적단핵감산다태성여고감유삼지(HTG)혈증지간적관계,탐토해기인변이여통풍병정진전적관계。방법측정중국산동지구300례통풍환자급300례건강인군중적ApoC-Ⅲ계동자적량개위점C-482T화T-455C적기인형화등위기인빈솔,동시측정량조인군적혈지수평,병대기진행비교。결과통풍조교대조조TG수평증고,차이유통계학의의(t=6.75,P<0.01)。455위점적CC、CT화TT기인형재통풍조화대조조중분포빈솔분별위23.0%、24.0%、53.0%화30.3%、13.3%、56.3%,차이유통계학의의(χ2=12.47,P<0.01);455위점등위기인 C 적분포빈솔재통풍조화대조조중49.5%화58.5%,차이유현저성통계학의의(χ2=9.78,P<0.01);482위점적CC、CT화TT기인형재통풍조화대조조중분포빈솔분별위28.0%、23.0%、49.0%화16.0%、33.7%、50.3%,차이유현저성통계학의의(χ2=15.89,P<0.01);482위점등위기인T적분포빈솔재통풍조화대조조중47.5%화58.8%,차이유통계학의의(χ2=15.47,P<0.01)。결론 ApoC-Ⅲ계동자적455화482위점적기인다태성여통풍환자적HTG발병유관。
Objective To study the relationship between single nucleotide polymorphism of promoter apolipoprotein C Ⅲ (ApoC-Ⅲ) and high triglyceride (HTG) of the gout patients, to explore the relationship between the gene mutation and gout disease. Methods Two promoter loci C-482T and T-455C genotype in ApoC-Ⅲ in 300 gout patients and 300 healthy control in Shandong province of China were determinated, blood lipid levels were also determined in the two groups, and then compared. Results The level of TG in gout group were higher than healthy control (t=6.75, P<0.01); there was significant difference between gout group and healthy control in loci 455, CC CT TT were 23.0%, 24.0%, 53.0% in gout group, and 30.3%, 13.3%, 56.3% in healthy control (χ2=12.47, P<0.01). The frequency distribution of gene C in loci 455 were respectively 49.5%and 58.5%in gout group and healthy control (χ2=15.89, P<0.01);there was significant difference between gout group and healthy control in loci 482, CC CT TT were 28.0%, 23.0%, 49.0%in gout group, and 16.0%, 33.7%, 50.3%in healthy control(χ2=15.89, P<0.01). The frequency distribution of gene C in loci 482 were respectively 47.5%and 58.8%in gout group and healthy control (χ2=15.47, P<0.01). Conclusion There were relationship between gene promoter 455 and 482 polymorphism in ApoC-Ⅲ and the pathogenesis of HTG in gout patients.
