Athyrium Mult identatum(Doll.) Ching%total amino acids%microwave method%orthogonal array experiments
目的:探讨利用微波提取猴腿蹄盖蕨总氨基酸的最佳工艺条件。方法:利用微波辅助法提取猴腿蹄盖蕨总氨基酸,采用紫外分光光度法进行猴腿总氨基酸的含量测定。结果:通过单因素与正交试验,得出猴腿蹄盖蕨总氨基酸的最佳提取工艺条件为:采用水提法,料液比为1:20(m/v),提取次数为2次,提取温度70-75℃,提取时间为25 min,微波提取功率为500 W,最佳工艺条件下,猴腿蹄盖蕨总氨基酸的平均提取率为19.063 mg/g。结论:微波辅助提取法适用于猴腿蹄盖蕨中总氨基酸的提取,具有快速、高效的特点。

Objective:To explore the optimum extraction technology of total amino acids from Athyrium Mult identatum(Doll.) Ching with microwave extraction method. Method:Microwave method was applied to extract the total amino acids from Athyrium Mult identatum (Doll.) Ching. and the concentration of total amino acids was determined by ultraviolet spectrophotometry. Results:With single factor and orthogonal array experiments, the optimum extraction condition of total amino acids from Athyrium Mult identatum (Doll.) Ching with microwave method using water as solvent was obtained as followed: the ratio between material and solvent (m/v), extraction times, extraction temperature, extraction time and microwave power were 1:20, 2 times, 70-75℃,25min and 500 W respectively. The average extracting efficiency of total amino acids could reach to 19.063mg/g under the optimum conditions. Conclusion:Microwave extraction method is fit for the extraction of total amino acids from Athyrium Mult identatum (Doll.) Ching , which was proved to be a fast and high efficient method .
