目的:观察冰片对食管癌细胞Eca109/DDP耐顺铂( DDP)的逆转作用,并探讨其机制。方法 MTT法检测冰片单用及与DDP联用对Eca109/DDP细胞增殖的影响,观察冰片对Eca109/DDP细胞耐DDP的逆转作用,计算耐药逆转指数;Western blot 法检测Eca109/DDP细胞P-糖蛋白( P-gp )的表达水平。结果0.195、0.390、0.780、1.560μg/mL冰片对Eca109/DDP细胞无明显毒性作用(P>0.05),但与1μg/mL的DDP联用能浓度依赖性增加DDP对Eca109/DDP细胞的毒性(P<0.05),降低DDP对Eca109/DDP细胞的半数抑制浓度(IC50)(P<0.05),耐药逆转指数高达5.31,并能下调Eca109/DDP细胞P-gp的表达水平(P<0.05)。结论冰片能逆转Eca109/DDP细胞对DDP的耐药,其作用与下调Eca109/DDP细胞P-gp的表达有关。

목적:관찰빙편대식관암세포Eca109/DDP내순박( DDP)적역전작용,병탐토기궤제。방법 MTT법검측빙편단용급여DDP련용대Eca109/DDP세포증식적영향,관찰빙편대Eca109/DDP세포내DDP적역전작용,계산내약역전지수;Western blot 법검측Eca109/DDP세포P-당단백( P-gp )적표체수평。결과0.195、0.390、0.780、1.560μg/mL빙편대Eca109/DDP세포무명현독성작용(P>0.05),단여1μg/mL적DDP련용능농도의뢰성증가DDP대Eca109/DDP세포적독성(P<0.05),강저DDP대Eca109/DDP세포적반수억제농도(IC50)(P<0.05),내약역전지수고체5.31,병능하조Eca109/DDP세포P-gp적표체수평(P<0.05)。결론빙편능역전Eca109/DDP세포대DDP적내약,기작용여하조Eca109/DDP세포P-gp적표체유관。
Objective To study the reversal effect and mechanism of borneol on cisplatin ( DDP) resistance in human esophageal carcinoma cell Eca 109/DDP.Methods The cytotoxic effect of borneol or DDP on Eca 109/DDP cells was measured by MTT and the drug -resistant reversal index (RI) was calculated.The expression of P-glycoprotein (P-gp) was detected by Western blot .Results MTT assay showed that borneol at the concentration no more than 1.560μg/mL had no obvious cytotoxicity to Eca 109/DDP cells ( P>0.05 ) , but it could significantly enhance the cytotoxicity of DDP (1 μg/mL) to Eca109/DDP cells in a concentration -dependent manner ( P <0.05), reduce IC50 of DDP on Eca109/DDP cells (P<0.05), and down-regulate the expression of P -gp of Eca109/DDP cells (P<0.05).RI of DDP was 5.31.Conclusion Borneol possesses a reversal effect on DDP resistance in Eca 109/DDP cells.The mechanism may be associated with the down -regulation of P-glycoprotein .
