目的：研究广藿香油抑制人前列腺癌细胞PC3增殖效应，探讨其诱导细胞凋亡作用及机制。方法MTT法观察广藿香油对细胞抑制作用；透射电镜观察细胞凋亡形态；流式细胞仪检测细胞周期变化；比色法检测caspase-3，caspase-9活性改变。结果 MTT法检测提示广藿香油体外抑制PC3细胞增殖，且抑制作用呈剂量-时间依赖性；透射电镜显示药物作用后细胞出现凋亡；流式细胞仪检测提示广藿香油作用使G0/G1期细胞比例上升，与对照组比较有统计学意义（P＜0.05）；caspase-3，caspase-9活性随药物作用剂量、时间增加逐渐递增。结论广藿香油体外抑制PC3细胞增殖，其抗肿瘤作用机制与阻断细胞周期、诱导细胞凋亡和激活caspase-3和caspase-9活性有关。
목적：연구엄곽향유억제인전렬선암세포PC3증식효응，탐토기유도세포조망작용급궤제。방법MTT법관찰엄곽향유대세포억제작용；투사전경관찰세포조망형태；류식세포의검측세포주기변화；비색법검측caspase-3，caspase-9활성개변。결과 MTT법검측제시엄곽향유체외억제PC3세포증식，차억제작용정제량-시간의뢰성；투사전경현시약물작용후세포출현조망；류식세포의검측제시엄곽향유작용사G0/G1기세포비례상승，여대조조비교유통계학의의（P＜0.05）；caspase-3，caspase-9활성수약물작용제량、시간증가축점체증。결론엄곽향유체외억제PC3세포증식，기항종류작용궤제여조단세포주기、유도세포조망화격활caspase-3화caspase-9활성유관。
Objective To investigate the mechanism of patchouli oil inhibiting the proliferation and inducing the apoptosis of human prostate cancer cells PC3. Methods The proliferation of cells was detected by MTT assay. The apoptosis morphology of cells was observed by transmission electron microscope; Cell cycle change was measured by flow cytometry;The activities of caspase-3 and caspase-9 were detected by colorimetric method. Results After treated with patchouli oil, apoptosis morphology of PC3 cells were found, the proliferation of PC3 cells was inhibited obviously with dose and time-dependent effects, the proportion of PC3 cells at G0/Gl phase was incresed(P<0.05), and the activities of caspase-3 and caspase-9 were enhanced with dose and time-dependent effects. Conclusion Patchouli oil inhibit the proliferation of PC3 cells which might be associated with G0/Gl phase arrest, cell apoptosis, and the activation of caspase-3 and caspase-9.
