目的:研究单味药洋金花细粉对帕金森病(PD)模型大鼠脑内多巴胺(DA)含量的影响及其作用机制。方法建立6-羟基多巴胺(6-OHDA)PD模型大鼠,以美多芭组、模型组、假手术组为对照,PD模型大鼠连续给予洋金花高、中、低剂量治疗3周后,将各组大鼠处死,立即取出脑组织并在冰盘上迅速分离出纹状体组织,应用高效液相色谱-质谱联用仪(TSQ)检测DA含量。多组间DA含量比较采用单因素方差分析,进一步两两比较采用LSD-t检验。结果模型对照组大鼠损毁侧纹状体DA含量明显降低,与假手术组比较差异极显著(P=0.00);给药3周后,各治疗组损毁侧纹状体DA含量均较模型组有所增加,洋金花高、中剂量组与模型对照组相比差异有统计学意义(P=0.00,P=0.03)。结论洋金花能增加PD模型大鼠纹状体DA的含量,补充黑质纹状体DA的不足,提高DA能神经元的活动,增强运动能力,从而改善PD模型大鼠的旋转行为。

목적:연구단미약양금화세분대파금삼병(PD)모형대서뇌내다파알(DA)함량적영향급기작용궤제。방법건립6-간기다파알(6-OHDA)PD모형대서,이미다파조、모형조、가수술조위대조,PD모형대서련속급여양금화고、중、저제량치료3주후,장각조대서처사,립즉취출뇌조직병재빙반상신속분리출문상체조직,응용고효액상색보-질보련용의(TSQ)검측DA함량。다조간DA함량비교채용단인소방차분석,진일보량량비교채용LSD-t검험。결과모형대조조대서손훼측문상체DA함량명현강저,여가수술조비교차이겁현저(P=0.00);급약3주후,각치료조손훼측문상체DA함량균교모형조유소증가,양금화고、중제량조여모형대조조상비차이유통계학의의(P=0.00,P=0.03)。결론양금화능증가PD모형대서문상체DA적함량,보충흑질문상체DA적불족,제고DA능신경원적활동,증강운동능력,종이개선PD모형대서적선전행위。
Objective To observe the impact of single herb Yangjinhua fine powder on the dopamine (DA) level in brain of parkinson disease (PD) model rats, and its mechanism thereof. Methods The rat model of PD was established by 6-hydroxydopamine (6-OHDA). The control groups included madopar group, model group and sham group. The high, medium, and low doses of Yangjinhua were given to PD model rats for three weeks. Rats were sacrificed and brains were removed immediately and quickly isolated in an ice tray to get striatal tissues. The DA levels of striatal tissues were detected by spectrometry (TSQ). Results Results of LSD-t test showed that the DA level of striatal tissues was significantly decreased in model group than that of sham group (P=0.00). The DA levels of striatal tissues were significantly increased in three Yangjinhua treatment groups than that of model group after three-week treatment (P=0.00,P=0.03).Conclusions The content of DA in striatum can be increased by Yangjinhua in PD model rats, which supply the deficiency of DA in striatum and enhance the activity of DA neurons, therefore, the rotation behavior was improved in PD model rats.
