采用cDNA末端快速扩增(rapid amplification of cDNA ends,RACE)技术,对纤毛虫原生动物扇形游仆虫(Eu-plotes vannus)谷胱甘肽过氧化物酶( GPx) cDNA特征进行分析。获得扇形游仆虫GPx cDNA全长序列,为672 bp,编码182个氨基酸,属硫氧还蛋白超家族,含有3个催化残基和3个特征性基序。氨基酸序列与其他纤毛虫物种同源性较高,亲缘关系较近。
採用cDNA末耑快速擴增(rapid amplification of cDNA ends,RACE)技術,對纖毛蟲原生動物扇形遊僕蟲(Eu-plotes vannus)穀胱甘肽過氧化物酶( GPx) cDNA特徵進行分析。穫得扇形遊僕蟲GPx cDNA全長序列,為672 bp,編碼182箇氨基痠,屬硫氧還蛋白超傢族,含有3箇催化殘基和3箇特徵性基序。氨基痠序列與其他纖毛蟲物種同源性較高,親緣關繫較近。
채용cDNA말단쾌속확증(rapid amplification of cDNA ends,RACE)기술,대섬모충원생동물선형유부충(Eu-plotes vannus)곡광감태과양화물매( GPx) cDNA특정진행분석。획득선형유부충GPx cDNA전장서렬,위672 bp,편마182개안기산,속류양환단백초가족,함유3개최화잔기화3개특정성기서。안기산서렬여기타섬모충물충동원성교고,친연관계교근。
Glutathione peroxidase ( GPx) has been widely used as a biomarker of environmental stress in ecotoxi-cology.In this study , GPx gene in Euplotes vannus was cloned and characterized .Degenerate primers were de-signed according to the glutathione peroxidase amino acid sequences in conservative area from similar species .The full-length cDNA was cloned from E.vannus using RACE-PCR.Results showed that the complete cDNA in E.van-nus is 672 bp encoding 182 amino acids and belongs to thioredoxin-like superfamily .Analyses of the derived amino acid sequence showed that GPx protein contains conservative structures , such as conserved catalytic residues and characteristic motifs , which is typical of glutathione peroxidase .Phylogenic analysis of amino acid sequences showed that E.vannus shares high homology and near affinity with other ciliate species .
