文章采用单因素实验方法对粒毛盘菌YM404产胞外黑色素(LEM404)液体培养基配方进行研究,得出其最佳配方如下:麦芽糖质量浓度为30 g/L、酵母膏质量浓度为6 g/L、L-DOPA浓度为1.25 mmol/L ,在该条件下获得LEM404的得率为3.45 g/L。采用葡聚糖凝胶柱G-100对LEM404进行分级分离,获得单一性组分LEM404-a。对LEM404-a进行质谱分析,表明其相对分子质量为432.18。配制6种LEM404-a质量分数不同的染发剂,其染发结果表明,以LEM404-a为原料配制的染发剂染色效果较好,但LEM404-a本身不能使白发着色。

The liquid fermentation medium components of Lachnum YM404 were determined by single factor experiments as follows :maltose 30 g/L ,yeast extract 6 g/L and L-DOPA 1.25 mmol/L .Un-der this condition ,the yield of LEM404 was 3.45 g/L .The extracted melanin (LEM404) was frac-tionated by Sephadex G-100 column ,and the homogeneous component LEM404-a was obtained .Ac-cording to mass spectrometry ,the relative molecular weight of LEM 404-a was 432.18 .Six kinds of hair dye with different LEM404-a content were prepared ,and the dyeing experiment suggested that the melanin hair dye with LEM404-a had significantly dyeing effect ,but LEM404-a could not make w hite hair hyperchromic by itself .
