Yishen Pinggan Decoction%Hypertension%Renal Damage%RAS
目的 1.观察益肾平肝方对血压的影响;2.观察益肾平肝方对自发性高血压大鼠(SHR)肾素血管紧张素系统(RAS)的影响.方法 以同龄雄性SHR和正常血压大鼠(Wistar-Kyoto rats,WKY)为研究对象,在10周龄时,SHR随机分为模型组(蒸馏水)、中药组(益肾平肝方)和西药组(洛汀新),WKY作为正常对照组(蒸馏水),每组8只,用药12周,检测实验第0、6、12周大鼠血压,实验12周末,检测大鼠血清及肾脏血管紧张素转换酶2(ACE2)和血管紧张素Ⅱ(AngⅡ)含量,所得结果进行统计分析.结果 12周时,与模型组比较发现,中药组、西药组大鼠血压明显降低(P< 0.01),中药组大鼠血清ACE2较WKY对照组及西药组明显减少(P<0.05),AngⅡ明显增加(P<0.05),与模型组无统计学差异(P>0.05),而二者在肾脏中的表达却不同,中药组大鼠肾脏ACE2较模型组明显增加(P<0.05),与西药组、WKY对照组无统计学差异(P>0.05),AngⅡ则明显减少(P<0.05),与WKY组无统计学差异(P>0.05).结论 益肾平肝方能降低SHR血压从而减轻肾脏损害,其作用可能与调节肾脏局部RAS的表达有关.

목적 1.관찰익신평간방대혈압적영향;2.관찰익신평간방대자발성고혈압대서(SHR)신소혈관긴장소계통(RAS)적영향.방법 이동령웅성SHR화정상혈압대서(Wistar-Kyoto rats,WKY)위연구대상,재10주령시,SHR수궤분위모형조(증류수)、중약조(익신평간방)화서약조(락정신),WKY작위정상대조조(증류수),매조8지,용약12주,검측실험제0、6、12주대서혈압,실험12주말,검측대서혈청급신장혈관긴장소전환매2(ACE2)화혈관긴장소Ⅱ(AngⅡ)함량,소득결과진행통계분석.결과 12주시,여모형조비교발현,중약조、서약조대서혈압명현강저(P< 0.01),중약조대서혈청ACE2교WKY대조조급서약조명현감소(P<0.05),AngⅡ명현증가(P<0.05),여모형조무통계학차이(P>0.05),이이자재신장중적표체각불동,중약조대서신장ACE2교모형조명현증가(P<0.05),여서약조、WKY대조조무통계학차이(P>0.05),AngⅡ칙명현감소(P<0.05),여WKY조무통계학차이(P>0.05).결론 익신평간방능강저SHR혈압종이감경신장손해,기작용가능여조절신장국부RAS적표체유관.
Objective To observe the effects of Yishen Pinggan Decoction on blood pressure,and to observe the effects of Yishen Pinggan Decoction on renin-angiotensin system (RAS) of spontaneously hypertensive rats (SHR).Methods Male spontaneously hypertensive rats (SHR) and normal Wistar-Kyoto rats(WKY) were studied at 10 weeks of age.SHR were randomly divided into model group (treated with distilled water),"Yishen Pinggan" group and lotensin group.And each group had eight rats.The WKY was used as the normal control group (treated with distilled water).They were continuously treated for 12 weeks.The blood pressure of rats was measured before experiment,6th week and 12th week after the experiment start.By the end of experiment on the 12th week,we detected the expression of ACE2,Ang Ⅱ in Serum and kidney on rat kidney,the results obtained by computer image analysis and statistical analysis.Results At 12 weekends,compared with model group we found that the blood pressure was significantly lower in TCM group and lotensin group (P < 0.01),the ACE2 of serum in TCM group was significantly reduced (P < 0.05) than in WKY rats and western medicine group,Ang Ⅱ then significantly increased (P < 0.05),with no significant difference between the model group (P > 0.05),while the expression in the kidney was different,the ACE2 of TCM group compared with the model group increased significantly (P < 0.05),and with western and WKY group,showing no significant difference (P > 0.05).Ang Ⅱ significantly reduced (P < 0.05),no significant difference found with WKY group (P > 0.05).Conclusion Yishen Pinggan Decoction can decrease blood pressure so as to reduce kidney damage,its effect may be related to regulating the expression of the relevant local kidney RAS.
