目的:探讨脑外伤合并骨折组与单纯骨折组骨折处骨痂组织中低氧诱导因子(HIF-1)的表达。方法对98只SPF级健康SD大鼠进行造模,其中A组(脑外伤合并骨折组)50例,B组(单纯骨折组)48例,造模成功后分别在1~4w每周周末剖取骨痂组织进行HE染色和免疫组化染色。利用高倍显微镜观察组织中4个互不重叠视野,计算细胞HIF-1阳性率,灰度值、光密度值、积分光密度值,观测细胞HIF-1的表达。结果 A组的各种指标均在第1w末达到高峰后开始换面下降,说明HIF-1在脑外伤合并骨折后增多,其后虽下降但任然活跃,加速骨折愈合。而单纯骨折组所有指标均无明显变化。结论脑外伤合并骨折组的骨痂组织中HIF-1的表达较单纯骨折组有明显增多。 HIF-1对骨折愈合有促进作用。这为日后临床治疗骨折愈合,提高治疗效率提供新方向和新靶点。

Objective The study set up traumatic brain injury associated with bone fracture rat model and bone fracture rat model. By HE staining and immunohistochemistry staining to detect the expression of HIF-1 in osteotylus. Methods:98 specified-pathogens free adult SD rats were divided into two groups randomly:(1)traumatic brain injury associated with fracture group(A,n=50);(2)fracture group(B,n=48). After set up model successful y,rats were kil ed respectively on 1st,2nd,3rd,4th week and intercepted osteotylus specimens from the fracture upper and lower about 5mm.Detect the expression of HIF-1 and analyes dates statictical y. Results: HIF-1 shows expression up-regulation in osteotylus specimens of A group, compared with B group, the results have statistical significance.It show that the healing speed of A group were much faster than B group.HIF-1 expression much large in 1st and decrease after that. While B group have no marked change on that. Conclusion:HIF-1 shows the expression up-regulatuion in cal us of fraeture sites in traumatic brain injury associated with bone fracture rat model. Compared with simple bone fracture rat model,the result show that HIF-1 can accelerated the healing of fracture.
