目的:评价流式荧光发光法在AFP、CEA、CA125、CA199定量测定中的可靠性。方法在Luminex200多功能流式点阵仪上评价了本法的精密度、准确度(回收实验)、检出限、线性和抗干扰性,并分析了本法与电化学发光法检测结果的相关性。结果批内变异系数均小于10%,批间变异每米均小于13%,回收率在89%~113%之间,灵敏度较好,线性良好(r>0.99),血红蛋白、胆红素、三酰甘油对测定结果无显著性干扰(P>0.05),与电化学发光法检测结果相关性良好(r>0.975)。结论本方法测定结果准确、可靠,操作简便、快速,适宜于临床检测推广。

Objective To evaluate the reliability of the quantitative determination of tumor markers AFP,CEA,CA125 and CA199 based on the flow cytometry fluorescence method.Methods The precision,accuracy,sensitivity,linearity and interference resistance of the kits were evaluated with the automated analyzed Luminex 200 as determination instrument,and the correlation of the results were analysis with that of electrochemiluminescence.Results The chemical character evaluation showed that the within-run and between-run coefficients of variation(CV)were less than 10% and 13%,respectively.The recovery rate was between 89%-113%,and the sensitivity was high.The results demonstrated excellent linearity(r>0.99).The assay had no significant in-terference by haemoglobin (Hb),bilirubin (Bil)and trig lyceride (TG)(P>0.05).The correlation between the evaluated kits and electrochemiluminescen was excellent (r>0.975).Conclusion The quantitative assay is safe,accurate and easy to operate,and it is worthy to extend in clinical application.
