PDCD4是一种重要的肿瘤抑制因子，它在肿瘤细胞中的表达下降或缺失，与肿瘤的治疗和预后具有相关性。本文主要阐述PDCD4在细胞氧化应激和DNA损伤中对DNA修复中的影响作用，并阐述PDCD4与肿瘤治疗过程中的耐药性相关进展。最新研究结果不仅为PDCD4的分子机制研究提供良好理论依据，而且为肿瘤个体化治疗提供参考，同时PDCD4也成为新药物开发的潜在靶点分子。

PDCD4 (Programmed cell death 4), an important tumor suppressor, often with decreased or absent expression in many tumor cells, is correlated with efficacy of chemotherapy and prognosis in certain tumor patients. This review focuses on the role of PDCD4 in cellular oxidative stress and DNA damage response, and the PDCD4 mediated molecular mechanisms underlying cell cycle control and drug resistance of tumor cells. Lastly, we put forward a new model of PDCD4-mediated sensitivity to paclitaxel by translational suppression of a group of proteins involved in mitotic exit. Latest advances of PDCD4 research not only provide a good theoretical basis for the personalized chemotherapy in practice, but also provide novel potential targets for drug development in the future.
