目的:观察不良因素应激对2型糖尿病动物模型的影响。方法雄性SD成年大鼠高脂饲料喂养4周,出现胰岛素抵抗后腹腔注射链脲佐菌素(STZ)30 mg/kg,制备糖尿病模型大鼠,将糖尿病模型大鼠随机分成两组,即糖尿病组和糖尿病应激组,正常大鼠随机分成两组,即正常对照组和正常应激组,糖尿病应激组和正常应激组大鼠进行束缚法应激,时间为每天9∶00 AM~3∶00 PM,每天束缚6 h,束缚期间禁水食,完成束缚应激后将大鼠放出,自由摄水食,应激时间共14 d,期间每天测量大鼠体重,进行水迷宫实验,第1天、第7天、第14天采血测量血糖、胰岛素、胰岛素敏感指数(ISI),进行统计学分析。结果与糖尿病模型组相比,糖尿病模型应激组大鼠血糖随应激实验的进行,血糖显箸升高(P<0.05),胰岛素敏感指数显著降低(P<0.05);糖尿病模型应激组逃避潜伏期显著延长(P<0.05),空间探索时间显著缩短(P<0.05)。结论高脂饮食结合STZ腹腔注射成功制备2型糖尿病大鼠模型;不良因素应激加重2型糖尿病大鼠模型的糖尿病临床症状。

목적:관찰불량인소응격대2형당뇨병동물모형적영향。방법웅성SD성년대서고지사료위양4주,출현이도소저항후복강주사련뇨좌균소(STZ)30 mg/kg,제비당뇨병모형대서,장당뇨병모형대서수궤분성량조,즉당뇨병조화당뇨병응격조,정상대서수궤분성량조,즉정상대조조화정상응격조,당뇨병응격조화정상응격조대서진행속박법응격,시간위매천9∶00 AM~3∶00 PM,매천속박6 h,속박기간금수식,완성속박응격후장대서방출,자유섭수식,응격시간공14 d,기간매천측량대서체중,진행수미궁실험,제1천、제7천、제14천채혈측량혈당、이도소、이도소민감지수(ISI),진행통계학분석。결과여당뇨병모형조상비,당뇨병모형응격조대서혈당수응격실험적진행,혈당현저승고(P<0.05),이도소민감지수현저강저(P<0.05);당뇨병모형응격조도피잠복기현저연장(P<0.05),공간탐색시간현저축단(P<0.05)。결론고지음식결합STZ복강주사성공제비2형당뇨병대서모형;불량인소응격가중2형당뇨병대서모형적당뇨병림상증상。
Objective To investigate effect of dys-psychological stress on the type 2 diabetes animal model. Methods Male SD adult rats fed high-fat diet 4 weeks, induced insulin resistance, given strep tozotocin (STZ, 30 mg/kg), Then they were randomly divided into 2 groups, the diabetes group and diabetes combined with dys-psychological stress group, the normal rats were also randomly divided into 2 groups, normal group and dys-psychological stress group. Morris water maze was used to evaluate the activity of rats. The body weight, blood glucose, insulin, insulin sensitivity index(ISI) were detected on the 1st, 7th, 14th after dys-psychological for statistical analysis. Results On the 1st, 4th, 7th after dys-psychological, blood glucose and ISI in diabetes combined with dys-psychological stress group significantly changed than diabetes group(P<0.05). The escape latency was significantly longer in the Morris water maze test, and the space exploration time was significantly shortened(P<0.05). Conclusion T2DM animal model could be induced by high-fat diet and STZ left-lower abdominal cavity injection, and the dys-psychological stress could make the clinical symptoms of type 2 diabetes worsen.
