研究正安野木瓜果实抗菌粗提物的优化提取工艺。以石油醚、氯仿、乙酸乙酯、丙酮、60%乙醇、95%乙醇和ddH2O等七种不同极性的溶剂为提取溶剂,研究溶剂因素对提取率和粗提物抗菌活性的影响。以60%乙醇为提取溶剂,单因素试验和正交试验考察液料比、温度及提取时间对野木瓜果实抗菌粗提物提取率的影响,确定合适的提取工艺。结果60%乙醇作为提取溶剂时,野木瓜果实抗菌粗提物提取率最高且有较好的抗菌活性。液料比和温度对提取率的影响较大;60%乙醇为提取溶剂时的最佳工艺为,液料比2.5∶1,浸提温度70℃,时间2 h。在该条件下,提取率为8.49%。液料比、温度和提取时间是影响抗菌粗提物提取率的主要因素。

Optimize extraction technology of antimicrobial crude extract from Stauntonvine fruit of Zheng-An. Stauntonvine fruit were taken with petroleum ether , chloroform , ethyl acetate , acetone , 60 % ethanol-solution, 95 % ethanol-solution and ddH2O as solvents, the effect of solvents on extraction ratio and antimicrobial activity were studied. Furthermore , 60 % ethanol-solution as extraction solvent , the effect on extraction ratio of ratio between solvents and material , temperature and time were investigate by means of single factor and orthogonal experiment and the optimum extraction condition was determined. Results 60 %ethanol-solution as extraction solvent , the extraction ratio of antimicrobial crude extract from Stauntonvine fruit was the highest, and the crude extract had good antimicrobial activity. The effect on extraction ratio of ratio between solvent and raw material and temperature were greater than that of extraction time. 60 %ethanol-solution as extraction solvent, the optimum technology was solvent/material:2.5∶1, temperature:70℃, extraction time: 2 h. The extraction ratio was 8.49 % at this condition. The effect on extraction ration of antimicrobial crude extract of extraction temperature , ratio between liquid and materials and extraction time were the main factors.
