目的:观察核桃青皮提取物对人食管癌细胞增殖的抑制作用。方法:用MTT法分别测定核桃青皮粗提物和核桃青皮提取物没食子酸、乙酸乙酯、紫杉叶素、7-D-芹菜糖-儿茶酚对食管癌细胞EC9706和KYSE150的增殖抑制率,并用Western Blot检测不同药物处理后食管癌细胞中细胞周期相关蛋白CyclinD1的蛋白表达。结果:核桃青皮粗提物、没食子酸和乙酸乙酯能抑制食管癌细胞的增殖,其作用呈明显剂量依赖性,当药物浓度增加至80μg/mL时,对KYSE150的抑制率分别为82.75%、90.51%、88.36%,对EC9706的抑制率分别为85.54%、87.21%、85.17%。与对照组相比,3种药物均会显著抑制KYSE150和EC9706细胞中CyclinD1的表达。结论:核桃青皮提取物对食管癌细胞系的增殖有较强的抑制作用,其机制可能与引起细胞周期阻滞有关。
目的:觀察覈桃青皮提取物對人食管癌細胞增殖的抑製作用。方法:用MTT法分彆測定覈桃青皮粗提物和覈桃青皮提取物沒食子痠、乙痠乙酯、紫杉葉素、7-D-芹菜糖-兒茶酚對食管癌細胞EC9706和KYSE150的增殖抑製率,併用Western Blot檢測不同藥物處理後食管癌細胞中細胞週期相關蛋白CyclinD1的蛋白錶達。結果:覈桃青皮粗提物、沒食子痠和乙痠乙酯能抑製食管癌細胞的增殖,其作用呈明顯劑量依賴性,噹藥物濃度增加至80μg/mL時,對KYSE150的抑製率分彆為82.75%、90.51%、88.36%,對EC9706的抑製率分彆為85.54%、87.21%、85.17%。與對照組相比,3種藥物均會顯著抑製KYSE150和EC9706細胞中CyclinD1的錶達。結論:覈桃青皮提取物對食管癌細胞繫的增殖有較彊的抑製作用,其機製可能與引起細胞週期阻滯有關。
목적:관찰핵도청피제취물대인식관암세포증식적억제작용。방법:용MTT법분별측정핵도청피조제물화핵도청피제취물몰식자산、을산을지、자삼협소、7-D-근채당-인다분대식관암세포EC9706화KYSE150적증식억제솔,병용Western Blot검측불동약물처리후식관암세포중세포주기상관단백CyclinD1적단백표체。결과:핵도청피조제물、몰식자산화을산을지능억제식관암세포적증식,기작용정명현제량의뢰성,당약물농도증가지80μg/mL시,대KYSE150적억제솔분별위82.75%、90.51%、88.36%,대EC9706적억제솔분별위85.54%、87.21%、85.17%。여대조조상비,3충약물균회현저억제KYSE150화EC9706세포중CyclinD1적표체。결론:핵도청피제취물대식관암세포계적증식유교강적억제작용,기궤제가능여인기세포주기조체유관。
Objective To observe the suppression effect of the extract of walnut green seedcase on the pro-liferation of esophageal cancer cell. Methods The suppression ratios of proliferation of esophageal cancer cell including EC9706 and KYSE150 caused by the extracts of walnut green seedcase, were measured using the MTT method respectively. The drugs derived from the extracts of walnut green seedcase were crude extract, gal-lic acid, EtoAC, taxifolin, and catechin-7-o-apiose. The expression of Cell cycle-related protein CyclinD1 in esophageal cancer cells was treated with different drugs by Western blotting. Results The crude extract, gal-lic acid and EtoAC had the suppression effect on the proliferation of esophageal cancer cells. The effects were dose-dependent significantly. When increasing the concentration to 80 μg/ mL, KYSE150 inhibition rates were 82.75%, 90.51%, and 88.36% respectively, and EC9706 inhibition rates were 85.54%,87.21%, and 85.17% re-spectively. Compared with the control group, these three drugs would significantly inhibited the expression of Cy-clinD1. Conclusion Some of the extracts of walnut green seedcase have the obvious suppression effect on the proliferation of esophageal cancer cells, which is associated with cell cycle retardation.
