Chuanxiong rhizoma%ligustilide%HPLC%processing method
目的:研究不同前处理方法对川芎药材中藁本内酯含量的影响。方法采用高效液相色谱法(HPLC)测定不同干燥方法和不同规格川芎药材中藁本内酯的含量。色谱条件:Kromasil C18(4.6 mm×250 mm,5μm),流动相为甲醇-水(77:23),流速1.0 mL/min,柱温25℃,检测波长324 nm,进样量10μL。结果藁本内酯在0.1623~2.4348μg范围内线性关系良好(r=0.9999);平均回收率100.3%,RSD=1.3%(n=6);不同干燥方法川芎药材中藁本内酯含量为2.02%~3.51%,川芎原药材和饮片中藁本内酯含量分别是1.89%与1.43%。结论自然晒干的川芎药材中藁本内酯含量高于低温烘干,川芎饮片中藁本内酯含量显著降低。

Objective To study the effect of different pretreatment methods on ligustilide in Chuanxiong rhizoma. Methods HPLC is used to determine ligustilide. The analysis was performed on a Kramasil C18 (4.6 mm×200 mm, 5μm) column. The mobile phase consisted of methanol-0.1%phosphoric acid (40:60). The flow rate was 1.0 mL?min-1. The detection wave1ength was 324 nm and column temperature was 25℃. Results The method displayed a good linearity within the concentration rang of 0.1623-2.4348μg (r=0.9999). The average recovery and RSD of ligustilide was 100.3%and 1.3%(n=6).The ligustilide in Chuanxiong rhizoma was 2.02%-3.51%. Conclution Sun-dried method is better than low temperature baking method. After processing treatment of Chuanxiong rhizoma, ligustilide content decreased significantly.
