为研究人参水提物(WEG)对SW480细胞体外增殖的抑制作用,采用倒置显微镜、MTT法观测SW480细胞的存活情况,并通过原子力显微镜获取各试验组细胞在人参水提物作用下细胞膜的平面/三维超微结构。结果表明:人参水提物能显著抑制SW480细胞增殖,其凋亡率与作用时间、剂量存在依赖关系(P<0.05),随着作用时间延长或剂量加大,对SW480细胞抑制作用呈增加趋势。原子力显微图像表明,正常SW480细胞呈圆形、细胞饱满,表面较光滑,而WEG处理组存活细胞皱缩,细胞表面的粗糙度较大,细胞膜存在一定的损伤。这说明WEG能显著抑制SW480细胞体外增殖,可能与WEG对SW480细胞膜的损伤有关。

위연구인삼수제물(WEG)대SW480세포체외증식적억제작용,채용도치현미경、MTT법관측SW480세포적존활정황,병통과원자력현미경획취각시험조세포재인삼수제물작용하세포막적평면/삼유초미결구。결과표명:인삼수제물능현저억제SW480세포증식,기조망솔여작용시간、제량존재의뢰관계(P<0.05),수착작용시간연장혹제량가대,대SW480세포억제작용정증가추세。원자력현미도상표명,정상SW480세포정원형、세포포만,표면교광활,이WEG처리조존활세포추축,세포표면적조조도교대,세포막존재일정적손상。저설명WEG능현저억제SW480세포체외증식,가능여WEG대SW480세포막적손상유관。
The mechanism of inhibition and apoptosis of WEG on the vitro colorectal cancer SW480 cell lines was studied .The effects of different doses of WEG on the proliferation of the SW480 cell lines were investigated by inverted microscope,MTT assay and atomic force microscope (AFM).The images from these three observation methods show that WEG can reduce SW480 cell viability in a dose-dependent and time-dependent manner (P<0.05).Those cells in the experimental groups were observed getting shrunk-en and round,membrane broken and surface rough,depending on the increasing dose .While the cells in the control group were round and full,the surfaces of the cells were smooth .The results show that WEG can inhibit the proliferation of SW480 cells significantly and this phenomenon may be caused by the injury of the cell membrane .
