目的:探讨供鼠NK细胞受体与受鼠MHCⅠ类分子匹配程度对骨髓移植小鼠造血重建的影响。方法分离30只C57BL/c小鼠的骨髓细胞和NK细胞,输给经过预处理的BALB/c和CB6F1小鼠,受鼠分为单纯辐照组、输入骨髓细胞组、输入骨髓细胞和NK细胞组,6只/组,观察不同时间点血常规、生存时间、病理组织学等指标变化,进行组间比较。结果输入骨髓细胞、输入骨髓细胞和NK细胞组小鼠均长期存活,未观察到GVHD病理学改变。移植后第14天时,输入骨髓细胞组和输入骨髓细胞和NK细胞组的BALB/c小鼠(MHCⅠ类分子全不相合)外周血WBC分别为(1.35±0.33)×109/L和(1.80±0.18)×109/L,差异有统计学意义,血小板计数分别为(79±19)×109/L和(117±13)×109/L,差异有统计学意义;输入骨髓细胞组和输入骨髓细胞和NK细胞组的CB6F1小鼠(MHCⅠ类分子半相合)外周血 WBC 分别为(1.52±0.26)×109/L 和(1.85±0.34)×109/L,差异无统计学意义,血小板计数分别为(90±12)×109/L 和(113±15)×109/L,差异具有统计学意义;BALB/c小鼠白细胞的改善略优于CB6F1小鼠。结论异源反应性NK细胞可以促进骨髓移植小鼠造血功能的恢复,并且这种作用似与NK细胞受体与受鼠MHC I类分子的匹配程度相关。

Objective To explore the effect of the ecxtent of matching between allo-NK cells receptors and major histocompatibility complex I molecules on the hematopoietic reconstitution after MHC haplotype mismatched BMT in mice. Methods The bone marrow cells and natural killer cells were separated from the C57BL/6 mice, and those bone marrow cells were transfused back into the γ-irradiation CB6F1 mice and BALB/c mice. GVHD and hematopoietic reconstitution were observed and evaluated by survival time, pathological changes and blood cells counting. Results Histopathology dis-played no GVHD in all the groups. For BALB/c mice,the leukocyte and platelet counts at day 14 after BMT were (1.35± 0.33)×109/L、(79±19)×109/L and (1.80±0.18)×109/L、(117±13)×109/L respectively for the infused bone marrow cells group and the infused bone marrow cells plus NK cells group. For CB6F1 mice, the leukocyte and platelet counts at day 14 after BMT were (1.52 ±0.26) ×109/L、(90 ±12) ×109/L and (1.85 ±0.34) ×109/L、(113 ±15) ×109/L respectively for the infused bone marrow cells group and the infused bone marrow cells plus NK cells group. Conclusion Donor- derived NK cells could promote hematopoietic reconstitution after BMT in mice, and the effect is closely related to the extent of matching between allo-NK cells receptors and major histocompatibility complex I molecules.
