目的:观察四妙勇安汤对缺氧人脐静脉内皮细胞( HUVEC )炎症因子白细胞介素-6( IL-6)、肿瘤坏死因子-α( TNF-α)及血管内皮生长因子( VEGF)水平的影响。方法将HUVEC细胞株进行传代培养后,将实验用细胞分为3组,正常组、模型组、四妙勇安汤组。正常组将正常的HUVEC细胞加入正常培养液培养;模型组将HUVEC细胞进行缺氧处理后加入正常培养液培养;四妙勇安汤组将HUVEC细胞进行缺氧处理后加入含10%四妙勇安汤醇沉药液的细胞培养液培养。用酶联免疫吸附法( ELISA )测定各组IL-6、TNF-α水平,用免疫组化法检测各组VEGF生成量的表达。结果模型组、四妙勇安汤组IL-6、TNF-α及VEGF水平与正常组比较差异均有统计学意义(P<0.01),IL-6、TNF-α上升,VEGF下降;但四妙勇安汤组IL-6、TNF-α上升水平及VEGF下降水平均小于模型组(P<0.01),四妙勇安汤可以改善缺氧HUVEC细胞中IL-6、TNF-α及VEGF水平变化。结论四妙勇安汤对缺氧HUVEC细胞能有效地抑制炎性因子的分泌,促进内皮细胞的增殖,具有抗炎及促进血管新生的作用,对损伤的血管内皮细胞具有一定的保护作用。

목적:관찰사묘용안탕대결양인제정맥내피세포( HUVEC )염증인자백세포개소-6( IL-6)、종류배사인자-α( TNF-α)급혈관내피생장인자( VEGF)수평적영향。방법장HUVEC세포주진행전대배양후,장실험용세포분위3조,정상조、모형조、사묘용안탕조。정상조장정상적HUVEC세포가입정상배양액배양;모형조장HUVEC세포진행결양처리후가입정상배양액배양;사묘용안탕조장HUVEC세포진행결양처리후가입함10%사묘용안탕순침약액적세포배양액배양。용매련면역흡부법( ELISA )측정각조IL-6、TNF-α수평,용면역조화법검측각조VEGF생성량적표체。결과모형조、사묘용안탕조IL-6、TNF-α급VEGF수평여정상조비교차이균유통계학의의(P<0.01),IL-6、TNF-α상승,VEGF하강;단사묘용안탕조IL-6、TNF-α상승수평급VEGF하강수평균소우모형조(P<0.01),사묘용안탕가이개선결양HUVEC세포중IL-6、TNF-α급VEGF수평변화。결론사묘용안탕대결양HUVEC세포능유효지억제염성인자적분비,촉진내피세포적증식,구유항염급촉진혈관신생적작용,대손상적혈관내피세포구유일정적보호작용。
Objective To investigate the effect of Simiaoyong'an decoction on IL-6, TNF-αand VEGF in human umbilical vein endothelial cells (HUVEC) with anoxia.Methods Human umbilical vein endothelial (HU-VEC) cell lines were subcultured .The cells were divided into three experimental groups , the normal group , model group, Simiaoyong'an decoction group .The normal HUVEC in the normal group were cultured in normal culture solu-tion.HUVEC in model group received anaerobic treatment and then were cultured by normal culture solution .HUVEC in Simiaoyong'an decoction group received anaerobic treatment and then were cultured by 10% Simiaoyong'an decoc-tion culture solution.The levels of IL-6, TNF-a and VEGF were measured by ELISA and immunohistochemistry in each group .Results There were significant differences between model group , Simiaoyong'an decoction group and the normal group on the levels of IL -6, TNF-a and VEGF ( P<0.01).The levels of IL -6 and TNF -αwas in-creased , and VEGF was decreased .But the increase of IL -6 and TNF-αand the decrease of VEGF in Simiaoyong'an decoction group was less than that in model group (P<0.01).Simiaoyong'an decoction can improve the levels of IL-6, TNF-a and VEGF in HUVEC with anoxia .Conclusion Simiaoyong'an decoction can effectively inhibit the se-cretion of inflammatory factors in human umbilical vein endothelial cells with anoxia , promote the proliferation of en-dothelial cells , have anti -inflammatory and angiogenic effects on vascular endothelial cell injury has a protective effect.
