目的:从受体后信号转导通路上重要调节因子环磷酸腺苷( cAMP)、蛋白激酶 A (PKA)、蛋白激酶 C(PKC)的角度,探讨慢性应激状态下大鼠下丘脑、胃肠道受体后信号转导的变化及电针百会、印堂、天枢穴改善抑郁症消化功能障碍的作用机理。方法将40只雄性 SD 大鼠随机分为正常组、模型组、对照组、电针组,每组10只。采用放射性同位素等方法,观察慢性应激模型大鼠下丘脑、胃窦及空肠组织的 cAMP 含量、PKA 及 PKC 活性的变化及电针印堂、百会和天枢穴的干预作用。结果模型组大鼠下丘脑、胃窦及空肠组织 cAMP 含量及 PKA 活性明显低于正常组,PKC 活性明显高于正常组,差异有统计学意义(P ﹤0.01);电针组及对照组 cAMP 含量及 PKA 活性明显均较模型组升高,PKC 活性较模型组明显降低,两组与模型组比较,差异有统计学意义(P ﹤0.01);电针组与对照组比较无明显差别。结论抑郁模型大鼠下丘脑、胃窦及空肠组织受体后 cAMP、PKA 信号通路下调,而 PKC 信号通路上调,电针改善抑郁症伴发的消化功能障碍可能与调节受体后 cAMP、PKA 及PKC 信号通路有关。

목적:종수체후신호전도통로상중요조절인자배린산선감( cAMP)、단백격매 A (PKA)、단백격매 C(PKC)적각도,탐토만성응격상태하대서하구뇌、위장도수체후신호전도적변화급전침백회、인당、천추혈개선억욱증소화공능장애적작용궤리。방법장40지웅성 SD 대서수궤분위정상조、모형조、대조조、전침조,매조10지。채용방사성동위소등방법,관찰만성응격모형대서하구뇌、위두급공장조직적 cAMP 함량、PKA 급 PKC 활성적변화급전침인당、백회화천추혈적간예작용。결과모형조대서하구뇌、위두급공장조직 cAMP 함량급 PKA 활성명현저우정상조,PKC 활성명현고우정상조,차이유통계학의의(P ﹤0.01);전침조급대조조 cAMP 함량급 PKA 활성명현균교모형조승고,PKC 활성교모형조명현강저,량조여모형조비교,차이유통계학의의(P ﹤0.01);전침조여대조조비교무명현차별。결론억욱모형대서하구뇌、위두급공장조직수체후 cAMP、PKA 신호통로하조,이 PKC 신호통로상조,전침개선억욱증반발적소화공능장애가능여조절수체후 cAMP、PKA 급PKC 신호통로유관。
Objective To explore the changes in the signal transduction pathway of hypothalamic and gastrointestinal receptors as well as the effect mechanism on digestive disturbance in depression treated with electroacupuncture(EA)at Baihui(GV20),Yintang(EX - HN3)and Tianshu(ST25)in terms of the im-portant regulatory factors such as cAMP,PKA and PKC. Methods Forty male SD rats were randomized into a normal group,a model group,a control group and an EA group,10 rats in each one. The radioisotope method was used to observe cAMP level,activity of PKA and PKC in hypothalamus,antrum and jejunum,as well as the intervention of EA at Yintang(EX - HN3),Baihui(GV20)and Tianshu(ST25). Results In the model group,cAMP level and PKA activity in hypothalamus,antrum and jejunum were lower apparently than those in the normal group in the rats,PKC activity was higher apparently than that in the normal group,indicating the significant difference(P ﹤ 0. 01). The cAMP level and PKA activity in EA group and the control group were all increased apparently than those in the model group,but PKC activity was reduced apparently than that in the model group. The differences were significant in comparison of these two groups with the model group(P ﹤ 0. 01). The difference was not significant between EA group and the control group. Conclusion The posterior signal transduction pathway,cAMP and PKA were down - regulated and PKC was up - regula-ted in hypothalamus,antrum and jejunum in the model rats of depression. EA treats digestive disturbance in depression,which is likely related to its regulatory effect on cAMP,PKA and PKC signal transduction.
