Baicalin%Lung injury%Lipopolysaccharide(LPS)%Mice
目的:研究黄芩苷预处理对气管滴注脂多糖(LPS)诱导的小鼠急性肺损伤(ALI)的保护作用。方法将40只小鼠随机分成正常组(Con 组)、模型组(Mod 组)、黄芩苷低剂量预处理组(Baicalin-L 组,25mg/kg)、黄芩苷高剂量预处理组(Baicalin-H 组,100mg/kg)及地塞米松组(Dex 组,5mg/kg),每组8只。采用气管内缓慢滴注5%的 LPS 溶液(5mg/kg)方法建立 ALI 模型。造模前4d,黄芩苷组分别向腹腔注射相应剂量的黄芩苷溶液,每天1次至造模当日;Dex 组,前3d 腹腔注射等容积的生理盐水,造模当日腹腔注射地塞米松溶液5mg/kg;Con 组和 Mod 组在相同时间点,腹腔注射等容积的生理盐水。处死小鼠,留取肺泡灌洗液(BALF),测定 BALF 中 IL-1β﹑IL-6和 TNF-α的浓度;取右上肺组织制作病理切片,HE 染色光镜观察。结果 HE 染色光镜下 Mod 组肺组织出现炎症反应重。Baicalin 组和 Dex 组肺组织病变较模型组有所改善。与Con 组比,实验各组大鼠 BALF 中 IL-1β﹑IL-6和 TNF-α含量均明显增加;Mod 组增幅最大,IL-1β﹑IL-6和 TNF-α含量显著高于 Dex 组和 Baicalin 预处理组;Baicalin-L 组显著高于 Dex 组和 Baicalin-H 组,差异均有统计学意义(P<0.01)。结论黄芩苷预处理可减少炎症介质释放,对 LPS 诱导的小鼠 ALI 有保护作用。

목적:연구황금감예처리대기관적주지다당(LPS)유도적소서급성폐손상(ALI)적보호작용。방법장40지소서수궤분성정상조(Con 조)、모형조(Mod 조)、황금감저제량예처리조(Baicalin-L 조,25mg/kg)、황금감고제량예처리조(Baicalin-H 조,100mg/kg)급지새미송조(Dex 조,5mg/kg),매조8지。채용기관내완만적주5%적 LPS 용액(5mg/kg)방법건립 ALI 모형。조모전4d,황금감조분별향복강주사상응제량적황금감용액,매천1차지조모당일;Dex 조,전3d 복강주사등용적적생리염수,조모당일복강주사지새미송용액5mg/kg;Con 조화 Mod 조재상동시간점,복강주사등용적적생리염수。처사소서,류취폐포관세액(BALF),측정 BALF 중 IL-1β﹑IL-6화 TNF-α적농도;취우상폐조직제작병리절편,HE 염색광경관찰。결과 HE 염색광경하 Mod 조폐조직출현염증반응중。Baicalin 조화 Dex 조폐조직병변교모형조유소개선。여Con 조비,실험각조대서 BALF 중 IL-1β﹑IL-6화 TNF-α함량균명현증가;Mod 조증폭최대,IL-1β﹑IL-6화 TNF-α함량현저고우 Dex 조화 Baicalin 예처리조;Baicalin-L 조현저고우 Dex 조화 Baicalin-H 조,차이균유통계학의의(P<0.01)。결론황금감예처리가감소염증개질석방,대 LPS 유도적소서 ALI 유보호작용。
Objective To study the effect of pretreatment with baicalin on acute lung injury induced by lipopolysaccha-ride(LPS) in mice. Methods 40 mice were randomly divided into control(Con) group, model(Mod) group,2 dose of baicalin groups (baicalin-L 25mg/kg and baicalin-H 100mg/kg),and dexamethasone group (Dex,5mg/kg), 8 mice in each group. 4 days before making the model,solution of corresponding dose baicalin was injected into the peritoneal cavity of mice in baicalin group,once a day; 3 days before making the model,equal volume of normal saline was intraperitoneally injected,at the day of making model,the dexamethasone solution (5mg/kg) was intraperitoneally administered to mice in Dex group; at the same time, intraperitoneal injection of normal saline was instilled in Con group and Mod group. ALI model was established by intratracheal instillation of LPS method. For making model,5 percent of LPS (5mg/kg) was slowly instilled into the trachea of mice in experi-mental groups,equal volume of saline was instilled in the Con group. 3 hours after instillation of LPS,the mice of all groups were sacrificed,and the brochoalveolar lavage fluid (BALF) was collected. Then the concentrations of interleukin 1β(IL-1β)、inter-leukin 6 (IL-6) and tumor necrosis factory (TNF-α) in BALF was measured by ELISA. Pathological section was made,and the pathological change was observed by light microscopic. Results The pathology observation of lung tissue of mice in the Mod group with light microscope showed that there was serious inflammation. The pathological changes of lung tissue in baicalin group and Dex group were lower than those of Mod group. Compared with the Con group,the levels of the IL-1β,IL-6 and TNF-α in BALF of mice in experimental groups were significantly higher,the levels of the IL-1β,IL-6 and TNF-α in Mod group were higher than those in Dex and baicalin groups. the difference were statistically significant (P<0.01). Conclusion Baicalin pretreatment can decrease inflammation and make some protective effect on ALI induced by LPS.
