目的:建立HPLC法测定兔血浆中柿叶总黄酮主要有效成分芦丁的方法,并用此法对柿叶总黄酮在兔体内的药动学进行研究。方法对6只雄性家兔耳缘静脉注射柿叶总黄酮,于不同时间取静脉血,用甲醇-冰醋酸(9∶1)沉淀蛋白,HPLC法测定芦丁的血药浓度;用3p87软件进行药动学参数计算。结果兔血浆芦丁浓度在0.1332~13.3200μg/ml范围内线性关系良好(r=0.9991);高、中、低三种浓度芦丁血浆样品日内平均RSD为7.13%,日间平均RSD为7.64%,平均回收率为(76.21±5.69)%。静脉注射给药后,芦丁在兔体内的药动学符合二室模型,半衰期t1/2(α)为(0.2865±0.0112)h,t1/2(β)为(1.7546±0.4451)h,AUC0→6为(8.0417±0.2169)μg/(h· ml), AUC0→∞为(8.1286±0.2293)μg/(h· ml)。结论所建方法灵敏、简便快速,专属性强,可满足柿叶总黄酮药动学研究的需要。
目的:建立HPLC法測定兔血漿中柿葉總黃酮主要有效成分蘆丁的方法,併用此法對柿葉總黃酮在兔體內的藥動學進行研究。方法對6隻雄性傢兔耳緣靜脈註射柿葉總黃酮,于不同時間取靜脈血,用甲醇-冰醋痠(9∶1)沉澱蛋白,HPLC法測定蘆丁的血藥濃度;用3p87軟件進行藥動學參數計算。結果兔血漿蘆丁濃度在0.1332~13.3200μg/ml範圍內線性關繫良好(r=0.9991);高、中、低三種濃度蘆丁血漿樣品日內平均RSD為7.13%,日間平均RSD為7.64%,平均迴收率為(76.21±5.69)%。靜脈註射給藥後,蘆丁在兔體內的藥動學符閤二室模型,半衰期t1/2(α)為(0.2865±0.0112)h,t1/2(β)為(1.7546±0.4451)h,AUC0→6為(8.0417±0.2169)μg/(h· ml), AUC0→∞為(8.1286±0.2293)μg/(h· ml)。結論所建方法靈敏、簡便快速,專屬性彊,可滿足柿葉總黃酮藥動學研究的需要。
목적:건립HPLC법측정토혈장중시협총황동주요유효성분호정적방법,병용차법대시협총황동재토체내적약동학진행연구。방법대6지웅성가토이연정맥주사시협총황동,우불동시간취정맥혈,용갑순-빙작산(9∶1)침정단백,HPLC법측정호정적혈약농도;용3p87연건진행약동학삼수계산。결과토혈장호정농도재0.1332~13.3200μg/ml범위내선성관계량호(r=0.9991);고、중、저삼충농도호정혈장양품일내평균RSD위7.13%,일간평균RSD위7.64%,평균회수솔위(76.21±5.69)%。정맥주사급약후,호정재토체내적약동학부합이실모형,반쇠기t1/2(α)위(0.2865±0.0112)h,t1/2(β)위(1.7546±0.4451)h,AUC0→6위(8.0417±0.2169)μg/(h· ml), AUC0→∞위(8.1286±0.2293)μg/(h· ml)。결론소건방법령민、간편쾌속,전속성강,가만족시협총황동약동학연구적수요。
Objective To develop a method for determining the plasma concentration of rutin from persimmon leaf total flavones in rabbits by using high performance liquid chromatography ( HPLC) and study its pharmacokinetics in rabbits in vivo .Methods Six male rabbits were given persimmon leaf total flavones by intravenous injection into the ear vein .Blood samples were collected from the ear vein at different time points;The plasma protein was precipitated directly with methanol-glacial acetic acid ( 9 ∶1 ) .The concentration of rutin in plasma was measured by HPLC .The pharmacokinetic parameters were estimated by pharmacokinetic program 3 p87 .Results The concentration of rutin in plasma achieved an excellent linear relationship in the range of 0.1332-13.3200 μg/ml(r=0.9991).The intra-day RSD was 7.13%,inter-day RSD was 7.64%,and the mean recovery was (76.21 ±5.69)% for rutin in plasma of three concentrations(high,middle and low concentrations).After single intravenous administration,the pharmacokinetic profile of rutin was fitted to the two-compartment model in rabbits.The main pharmacokinetics parameters were as follows:t1/2(α) was (0.2865 ±0.0112) hour,t1/2(β) was(1.7546 ±0.4451) hours,AUC0→6 was (8.0417 ±0.2169)μg/(h· ml), AUC0→∞ was (8.1286 ±0.2293)μg/(h· ml).Conclusion This HPLC method is sensitive,simple and efficient with excellent specificity ,which can be applied to pharmacokinetic study on persimmon leaf total flavones .
