目的:研究人eag相关基因(HERG)蛋白表达对胃癌细胞凋亡及细胞周期分布的影响。方法构建HERG-siRNA载体;将构建的载体转染胃癌细胞SGC7901,氨基糖苷类抗生素G418进行转染细胞的稳定筛选并克隆化,从蛋白和电流水平鉴定转染细胞。采用流式细胞仪检测胃癌细胞凋亡情况;采用透射电子显微镜技术观察胃癌细胞超微结构的变化;采用流式细胞仪检测胃癌细胞细胞周期分布的变化。结果HERG-siRNA可以阻断胃癌细胞中的HERG电流,可以诱导胃癌细胞出现凋亡;在透射电子显微镜下观察到典型的凋亡细胞的形态学改变;HERG-siRNA可以阻止胃癌细胞由G1期进入S期。结论HERG-siRNA可抑制HERG蛋白及相应钾电流,从而促进胃癌细胞凋亡,抑制胃癌细胞G1/S期转化,具有潜在的抗癌作用。

목적:연구인eag상관기인(HERG)단백표체대위암세포조망급세포주기분포적영향。방법구건HERG-siRNA재체;장구건적재체전염위암세포SGC7901,안기당감류항생소G418진행전염세포적은정사선병극륭화,종단백화전류수평감정전염세포。채용류식세포의검측위암세포조망정황;채용투사전자현미경기술관찰위암세포초미결구적변화;채용류식세포의검측위암세포세포주기분포적변화。결과HERG-siRNA가이조단위암세포중적HERG전류,가이유도위암세포출현조망;재투사전자현미경하관찰도전형적조망세포적형태학개변;HERG-siRNA가이조지위암세포유G1기진입S기。결론HERG-siRNA가억제HERG단백급상응갑전류,종이촉진위암세포조망,억제위암세포G1/S기전화,구유잠재적항암작용。
Objective To explore the effects of human ether-a-go-go-related gene ( HERG ) protein on cell cycle and apoptosis of gastric cancer cells .Methods HERG-siRNA vector was constructed and transfected into gastric cancer cell SGC7901 ,G418 was applied to screening and cloning the transfected cells which were verified with HERG current and protein .Flow cytometry was used to assess the apoptosis of gastric cancer cells;Transmission electron microscope was used to observe the ultrastructure of gastric cancer cells ;The cell cycle distribution of gastric cancer cells was investigated by flow cytometry .Results HERG-siRNA eliminated the HERG current in gastric cancer cells and induced apoptosis in gastric cancer cells;There were typical changes of apoptosis under transmission electron microscope;HERG-siRNA inhibited gastric cancer cells entering into S phage from G 1 phage.Conclusion HERG-siRNA can inhibit HERG protein and potassium current as well as the progression of G 1/S of gastric cancer cells ,and induce apoptosis of gastric cancer cells .HERG -siRNA has potential anti-cancer effects on gastric cancer cells .
