目的研究白芍总苷(TGP)对内毒素性心肌炎大鼠心肌组织中炎症因子TNF-α、IL-1β、NF-κB表达的影响。方法通过尾静脉注入脂多糖(LPS)建立内毒素性心肌炎大鼠模型,分别以低、中、高剂量的TGP进行干预,光镜观察大鼠心肌组织的病理学变化,免疫组化检测心肌组织TNF-α、IL-1β、NF-κB的表达水平。结果光镜观察发现模型组大鼠心肌组织炎症反应明显,较多炎症细胞浸润;TGP中、高剂量组大鼠心肌组织炎症反应较轻,有少量炎症细胞浸润;低剂量组大鼠心肌组织炎症坏死程度介于模型组和中剂量组之间。免疫组化检测提示TGP中、高剂量组TNF-α、IL-1β、NF-κB的表达明显减少,与模型组比较均有统计学差异(均P<0.01);TGP低剂量组的TNF-α、IL-1β表达与模型组均无统计学差异(均P>0.05),而NF-κB的表达与模型组有统计学差异(P<0.05)。结论 TGP能有效抑制内毒素性心肌炎大鼠心肌组织炎症反应,减轻心肌损伤,在中、高剂量时效果明显。

목적연구백작총감(TGP)대내독소성심기염대서심기조직중염증인자TNF-α、IL-1β、NF-κB표체적영향。방법통과미정맥주입지다당(LPS)건립내독소성심기염대서모형,분별이저、중、고제량적TGP진행간예,광경관찰대서심기조직적병이학변화,면역조화검측심기조직TNF-α、IL-1β、NF-κB적표체수평。결과광경관찰발현모형조대서심기조직염증반응명현,교다염증세포침윤;TGP중、고제량조대서심기조직염증반응교경,유소량염증세포침윤;저제량조대서심기조직염증배사정도개우모형조화중제량조지간。면역조화검측제시TGP중、고제량조TNF-α、IL-1β、NF-κB적표체명현감소,여모형조비교균유통계학차이(균P<0.01);TGP저제량조적TNF-α、IL-1β표체여모형조균무통계학차이(균P>0.05),이NF-κB적표체여모형조유통계학차이(P<0.05)。결론 TGP능유효억제내독소성심기염대서심기조직염증반응,감경심기손상,재중、고제량시효과명현。
Objective To investigate whether total glucosides of paeony (TGP) could inhibit expression of inflammatory factors in myocardial tissue of rats with myocarditis. Methods Myocarditis was induced by injection of endotoxin LPS through the tail vein in rats; then low, middle or high doses of TGP were administered by gastric perfusion respectively. Histopathological changes were examined by HE staining;the expression of TNF- α, IL- 1βand NF- κB in myocardial tissue was detected by im-munohistochemistry. Results Histopathology demonstrated marked inflammation in cardiac tissue with infiltration of inflammatory cells. TGP with middle and high dose al eviated inflammation in cardiac tissue. Expression of TNF- α, IL- 1βand NF- κB was up- graded in rats with myocarditis, which was attenuated in middle and high dose- TGP groups(P<0.01). Conclusion TGP with middle or high dose can effectively inhibit inflammation in cardiac tissue of rats with endotoxin- induced myocarditis.
