通过4种具有代表性的造模方法诱导高脂血症大鼠模型,筛选出理想的高脂血症造模方法。将健康SPF级雄性SD大鼠50只随机平均分为空白组、模型组Ⅰ~Ⅳ。空白组正常基础饲料喂养;各模型组分别采用高脂饲料喂养和高脂乳剂灌胃法诱导造模,模型组Ⅰ:基础饲料59%+食用油15%+蛋黄粉10%+蔗糖10%+胆固醇6%;模型组Ⅱ:基础饲料56.8%+食用油15%+蛋黄粉10%+蔗糖10%+胆固醇6%+胆酸钠2%+丙基硫氧嘧啶0.2%;模型组Ⅲ:正常饲料喂养配合高脂乳剂灌胃(高脂乳剂配方:吐温60%+食用油15%+胆固醇6%+胆酸钠2%+丙基硫氧嘧啶0.2%);模型组Ⅳ:一次性腹腔注射维生素D360万单位/kg后,饲养与灌胃方法同模型组Ⅲ。在造模10、20、40 d时经大鼠眼眶静脉丛取血,提取血清,检测血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)并计算肝指数水平。其中模型组Ⅱ、模型组Ⅲ和模型组Ⅳ均出现血脂代谢紊乱,肝指数明显升高(P <0.05),提示高脂血症模型构建成功,且模型组Ⅳ造模周期明显较模型组Ⅱ、Ⅲ缩短。

To screenr an ideal method which can build hyperlipidemia rat model out of different modeling methods,we divided 50 rats stochastically as the blank group,the model groupⅠ,the model groupⅡ,the model groupⅢ and the model groupⅣ,10 rats in each group.Except the blank group,four different mod-eling methods were used to build hyperlipidemia rat models.GroupⅠ:basal diet(5 9%)+ edible oil(1 5%)+ yolk powder(10%)+ sucrose (10%)+ cholesterol(6%);Group Ⅱ:basal diet(56.8%)+ edible oil (15%)+ yolk powder(10%)+ sucrose (10%)+ cholesterol(6%)+ sodium cholate(2%)+ propyl-thiouracil(0.2%);Group Ⅲ:normal diet respectively with intragastric administration of fat emulsion (fat emulsion formulation:Tween(60%)+ edible oil(15%)+ cholesterol(6%)+ sodium cholate(2%)+propylthiouracil(0.2%));Group Ⅳ:before intragastric administration of fat emulsion-the same as model group Ⅲ,give an intraperitoneal injection of vitamin D3 60 000 U/kg.We took the orbital venous plexus blood of all the rats in 10 d,20 d and 40 d,isolated sera then detected the levels of TC,TG,HDL-C and LDL-C by ELISA,and detected the levels of body weights and the liver indexes of rats in each group.The results showed that rats became hyperlipoidemic in groups Ⅱ,Ⅲ andⅣ,and the hyperlipidemia model was formed successfully.Compared with model group Ⅱ and group Ⅲ ,the modeling time of group Ⅳ was sig-nificantly reduced.
