为分析感染高原鼠兔的皮蝇蛆分子分类地位,通过聚合酶链反应(PCR)对6株感染高原鼠兔的皮蝇蛆的线粒体CO1基因片段进行扩增、克隆和测序,并将其序列与 GenBank中的皮蝇科、胃蝇科、狂蝇科的序列进行了比对分析,构建了遗传进化树。结果显示,扩增出的CO1基因片段为688 bp,感染高原鼠兔的皮蝇蛆与皮蝇科的皮蝇蛆遗传距离更近,位于一个大的进化树分支上,其基因片段同源性为81.2%~86.6%;与胃蝇科的蝇蛆基因片段同源性为77.8%~80.4%,与羊狂蝇基因片段同源性为79.4%~79.8%。结果表明,试验中的6株感染高原鼠兔的皮蝇蛆之间的基因片段同源性98.1%~100%,为皮蝇科的皮蝇蛆。

위분석감염고원서토적피승저분자분류지위,통과취합매련반응(PCR)대6주감염고원서토적피승저적선립체CO1기인편단진행확증、극륭화측서,병장기서렬여 GenBank중적피승과、위승과、광승과적서렬진행료비대분석,구건료유전진화수。결과현시,확증출적CO1기인편단위688 bp,감염고원서토적피승저여피승과적피승저유전거리경근,위우일개대적진화수분지상,기기인편단동원성위81.2%~86.6%;여위승과적승저기인편단동원성위77.8%~80.4%,여양광승기인편단동원성위79.4%~79.8%。결과표명,시험중적6주감염고원서토적피승저지간적기인편단동원성98.1%~100%,위피승과적피승저。
The obj ectives of this study were to analyze of molecular taxonomic positions of Hypoderma lar-vae in the plateau pika.The partial sequence of cytochrome oxidase 1 gene (CO1)from 6 strains of Hypo-derma larvae in plateau pika were amplified,cloned,sequenced and compared with Hypodermatidae,Gaster-ophilidae,Oestridae in GenBank,and phylogenetic analyses were also constructed.The results showed that Hypoderma larvae in the plateau pika and other Hypoderma larvae have a closer genetic relationship, clustered in one large branch of the phylogenetic tree,with 81.2%-86.6% in genetic homology ;and the sequence identities is 77.8%-80.4% with Gasterophilidae larvae,and 79.4%-79.8% with Oestrus ovis . The genetic similarity of CO1 fragments between 6 Hypoderma larvae in plateau pika is 98.1%-100%,be-longing to Hypodermatidae.
