扩张床吸附是一种新型的集固液分离、浓缩、纯化于一体的层析技术,其核心为经过特殊设计的吸附介质。比较了Streamline SP、 Streamline SP XL和 Fastline SP三种常用扩张床介质的理化性质和床层特性,并以乳铁蛋白为模型蛋白,从静态吸附、吸附动力学和动态吸附三方面比较了不同介质的吸附特性。结果表明,介质颗粒小,吸附平衡快;介质密度大,床层稳定性高,适用流速大,过程处理能力高,研究的结果为扩张床吸附介质的理性设计提供指导。
擴張床吸附是一種新型的集固液分離、濃縮、純化于一體的層析技術,其覈心為經過特殊設計的吸附介質。比較瞭Streamline SP、 Streamline SP XL和 Fastline SP三種常用擴張床介質的理化性質和床層特性,併以乳鐵蛋白為模型蛋白,從靜態吸附、吸附動力學和動態吸附三方麵比較瞭不同介質的吸附特性。結果錶明,介質顆粒小,吸附平衡快;介質密度大,床層穩定性高,適用流速大,過程處理能力高,研究的結果為擴張床吸附介質的理性設計提供指導。

Expanded bed adsorption (EBA) is a challenge bioseparation approach, which integrates the solid-liquid separation, concentration and primary purification into a unit operation. The key to the EBA is the specially-designed adsorbents. In the present work, the physical and chemical properties of three widely-used EBA adsorbents named Streamline SP, Streamline SP XL and Fastline SP were compared. The expansion characteristic and liquid mixing property of the beds formed by using above three adsorbents separately were compared too. With lactoferrin as the model protein, the adsorption properties including adsorption isotherms, adsorption kinetics and breakthrough curves of the three adsorbents were investigated. The results indicate that the adsorbent with small size can come to adsorption equilibrium faster and the adsorbent with high density could improve the bed stability and allow higher flow rate, which enhances the treating ability of the bed. This work would provide useful information for the design of new EBA adsorbents.
