Acetaminophen succinate%Pharmacokinetics%HPLC
目的:研究肌肉注射对乙酰氨基酚琥珀酸酯在健康家兔体内的药代动力学。方法:采用RP-HPLC法测定6只成年健康家兔肌肉注射对乙酰氨基酚琥珀酸酯后不同时间点血浆中对乙酰氨基酚的浓度,应用DAS 2.0药动学软件处理血药浓度数据,拟合对乙酰氨基酚琥珀酸酯在家兔体内吸收模型,绘制药时曲线,计算药代动力学参数。结果:对乙酰氨基酚琥珀酸酯在家兔体内代谢符合一级吸收二室模型(权重系数=1/C2),主要药动学参数:Tmax=0.25h、Cmax=95.44mg/L、t1/2(α)=0.73h、t1/2(β)=2.70h、AUC=82.44mg/L*h、Vd=45.65L/kg。结论:与肌肉注射对乙酰氨基酚相比,家兔肌肉注射乙酰氨基酚琥珀酸酯的Tmax、t1/2(β)均缩短,表明乙酰氨基酚琥珀酸酯在体内起效迅速且消除较快。
目的:研究肌肉註射對乙酰氨基酚琥珀痠酯在健康傢兔體內的藥代動力學。方法:採用RP-HPLC法測定6隻成年健康傢兔肌肉註射對乙酰氨基酚琥珀痠酯後不同時間點血漿中對乙酰氨基酚的濃度,應用DAS 2.0藥動學軟件處理血藥濃度數據,擬閤對乙酰氨基酚琥珀痠酯在傢兔體內吸收模型,繪製藥時麯線,計算藥代動力學參數。結果:對乙酰氨基酚琥珀痠酯在傢兔體內代謝符閤一級吸收二室模型(權重繫數=1/C2),主要藥動學參數:Tmax=0.25h、Cmax=95.44mg/L、t1/2(α)=0.73h、t1/2(β)=2.70h、AUC=82.44mg/L*h、Vd=45.65L/kg。結論:與肌肉註射對乙酰氨基酚相比,傢兔肌肉註射乙酰氨基酚琥珀痠酯的Tmax、t1/2(β)均縮短,錶明乙酰氨基酚琥珀痠酯在體內起效迅速且消除較快。
목적:연구기육주사대을선안기분호박산지재건강가토체내적약대동역학。방법:채용RP-HPLC법측정6지성년건강가토기육주사대을선안기분호박산지후불동시간점혈장중대을선안기분적농도,응용DAS 2.0약동학연건처리혈약농도수거,의합대을선안기분호박산지재가토체내흡수모형,회제약시곡선,계산약대동역학삼수。결과:대을선안기분호박산지재가토체내대사부합일급흡수이실모형(권중계수=1/C2),주요약동학삼수:Tmax=0.25h、Cmax=95.44mg/L、t1/2(α)=0.73h、t1/2(β)=2.70h、AUC=82.44mg/L*h、Vd=45.65L/kg。결론:여기육주사대을선안기분상비,가토기육주사을선안기분호박산지적Tmax、t1/2(β)균축단,표명을선안기분호박산지재체내기효신속차소제교쾌。
Objective:To study the pharmacokinetics of acetaminophen succinate in healthy rabbit after intramuscular injection.Methods:RP-HPLC was used to determine the acetaminophen concentration of 6 adult healthy rabbits at different time after injecting acetaminophen succinate. Then the pharmacokinetic parameters were disposed by DAS 2.0 pharmacokinetic software.Results:The concentration-time curve of acetaminophen succinate in rabbit was single compartment model (weight coefficient=1/C2). The main pharmacokinetic parameters was:Tmax=0.25h, Cmax=95.44mg/L, t1/2(α)=0.73h, t1/2(β)=2.70h, AUC=82.44 mg/L*h, Vd=45.65L/kg.Conclusions:Compared with muscular injection of acetaminophen, the Tmax and t1/2(β) of muscular injection of acetaminophen succinate shorten obviously, which indicates that acetaminophensuccinateonsetquicklyandremoverapidly in vivo.
