通过分光光度法建立了一套简便、准确检测云南萝芙木中利血平的新方法并确定其最适反应体系。该方法基于利血平芳香环结构与2,4-二硝基-6-溴苯胺重氮化盐偶合反应,其红褐色偶合产物与利血平浓度呈正相关。结果表明,该方法反应灵敏、专一,产物稳定性好。反应体系最适检测波长510 nm,最适反应温度和时间分别为40℃、10 min,最适反应摩尔比为1︰1,反应终止0.5 h检测适宜,体系反应过程宜遮光。利血平在0~60μg/mL遵从比尔定律。通过对萝芙木甲醇浸提物的检测结果表明,该方法与HPLC法没有明显差异,有效性高,适合萝芙木原料及产品中利血平的快速检测及其质量监控。

A simple and accurate method for detection of reserpine in Rauvolfia yunnanensis was established using spectrophotometer , and the optimum reaction system was established .The method was based on the aromatic ring of reserpine could couple with 2, 4-dinitro-6-bromine aniline diazotized salt , and the concentration of coupling product with red-brown color had positive correlation with the concentration of reserpine .The results verified that the development method was a sensitive and specific determination method with stable reaction product .Analysis showed that the reaction to be completed in 10 min at 40℃temperature , the maximum absorption was obtained at 510 nm when drug to reagent mole ratio was 1︰1 to form the azo adduct .It was appropriate to detect the absor-bance after terminated reaction about 0.5 hour and the sample should wrapped during the whole process .The con-centration of reserpine followed Bill ’ s law in 0~60 ug/mL with good reproducible and specificity .The method was suitable to assay the reserpine concentration and quality control for Rauvolfia yunnanensis raw materials or products .
