Zhengyuan Prescription%cyclophosphamide%white blood cell%myelosuppression
目的:探讨正源方对环磷酰胺(CTX)致豚鼠骨髓抑制的保护作用。方法将豚鼠随机分为对照组、模型组、rhG-CSF组(20μg·kg -1)、正源方低、中、高剂量组(2.00,4.00,8.00g·kg -1)。正源方各给药组及rhG-CSF组在造模前提前给药3d,其余组给予同等剂量的生理盐水,连续给药18 d。除对照组外,实验第4天各组肌肉注射CTX(64 mg · kg -1),连续3 d。于造模后第8天观察外周血象。实验结束后测定心、肝、脾、肺、肾等脏器指数,在显微镜下观察骨髓涂片的变化。结果与对照组比较,模型组豚鼠的外周血白细胞、红细胞、淋巴细胞、中性粒细胞计数、血红蛋白含量均不同程度地降低( P<0.01或 P<0.05),骨髓涂片显示,骨髓巨核细胞受到不同程度地破坏;与模型组比较,各给药组的白细胞计数均显著上升(P<0.01),以rhG-CSF组提升白细胞、中性粒细胞计数最为显著(P<0.01);其中正源方中剂量组的白细胞、红细胞、淋巴细胞、中性粒细胞计数、血红蛋白含量均显著上升(P<0.01或 P<0.05);各给药组豚鼠的心、肝、脾、肺、肾指数并无显著变化(P>0.05);骨髓涂片显示,正源方中剂量组对环磷酰胺抑制的骨髓有核细胞和巨核细胞系统修复作用最好。结论正源方能够促进环磷酰胺所致豚鼠骨髓抑制外周血的恢复和骨髓细胞的再生,对豚鼠的骨髓造血系统有一定的保护作用。

목적:탐토정원방대배린선알(CTX)치돈서골수억제적보호작용。방법장돈서수궤분위대조조、모형조、rhG-CSF조(20μg·kg -1)、정원방저、중、고제량조(2.00,4.00,8.00g·kg -1)。정원방각급약조급rhG-CSF조재조모전제전급약3d,기여조급여동등제량적생리염수,련속급약18 d。제대조조외,실험제4천각조기육주사CTX(64 mg · kg -1),련속3 d。우조모후제8천관찰외주혈상。실험결속후측정심、간、비、폐、신등장기지수,재현미경하관찰골수도편적변화。결과여대조조비교,모형조돈서적외주혈백세포、홍세포、림파세포、중성립세포계수、혈홍단백함량균불동정도지강저( P<0.01혹 P<0.05),골수도편현시,골수거핵세포수도불동정도지파배;여모형조비교,각급약조적백세포계수균현저상승(P<0.01),이rhG-CSF조제승백세포、중성립세포계수최위현저(P<0.01);기중정원방중제량조적백세포、홍세포、림파세포、중성립세포계수、혈홍단백함량균현저상승(P<0.01혹 P<0.05);각급약조돈서적심、간、비、폐、신지수병무현저변화(P>0.05);골수도편현시,정원방중제량조대배린선알억제적골수유핵세포화거핵세포계통수복작용최호。결론정원방능구촉진배린선알소치돈서골수억제외주혈적회복화골수세포적재생,대돈서적골수조혈계통유일정적보호작용。
Objective To investigate the effects of Zhengyuan Prescription on the myelosuppression of guinea pigs induced by cyclo-phosphamide(CTX) .Methods Guinea pigs were randomly divided into control group ,model group ,rhG-CSF group (20 μg · kg -1 ) ,and three groups each containing low (2 .00 g · kg -1 ) ,medium(4 .00 g · kg -1 ) ,high(8 .00 g · kg -1 ) dose of Zhengyuan Prescription .Zhengyuan Prescription groups and rhG-CSF group were administered for 3 days before modeling ,while the remai-ning groups received physiological saline with equal volumes ,and the duration of the experiment was 18 days .Except for the con-trol group ,the other guinea pigs were induced by CTX (64 mg · kg -1 ,im ,for 3 days) on the 4th day .Peripheral blood was ob-served at the 8th day after modeling .After the experiment ,the organ indexes of heart ,liver ,spleen ,lung ,and kidney were meas-ured and the changes of bone marrow smears were observed under the microscope .Results Compared with the control group ,the number of peripheral white blood cell(WBC) ,red blood cell(RBC) ,lymphocyte ,neutrophil and hemoglobin content of the model group decreased significantly (P<0 .01 or P<0 .05) .Furthermore ,the results of bone marrow smear revealed that bone marrow megakaryocytes were damaged in varying degrees .Compared with the model group ,WBC counts of the treatment groups were in-creased significantly(P<0 .01) ,especially for the group of rhG-CSF ,the WBC and neutrophil counts were enhanced most remark-ably(P<0 .01) .The blood cells counts of guinea pigs in the group with medium dose of Zhengyuan Prescription were increased significantly(P<0 .01 or P<0 .05) ,while the organ indexes of heart ,liver ,spleen ,lung ,and kidney had no significant changes(P>0 .05) .The results of bone marrow smear showed that the group of Zhengyuan Prescription with medium dose had a obvious therapeutic effect on the bone marrow nucleated cells and megakaryocytic cells system which was suppressed by CTX .Conclusion The recovery of peripheral blood and regeneration of bone marrow cells in myelosuppressed guinea pigs induced by CTX can be promoted by Zhengyuan Prescription which had a protective effect on the bone marrow hematopoietic system .
