目的:优化血浆中人凝血因子Ⅷ提取工艺条件。方法以人血浆冷沉淀为原料,人凝血因子Ⅷ的比活性回收率作为评价指标,采用单因素试验,对铝胶的加量,铝胶吸附时的pH值、酸沉淀时的pH值等进行研究,优选提取工艺。结果加入冷沉淀重量12%的铝胶,并将pH值调节至7.1进行吸附,上清液中人凝血因子Ⅷ比活性可达9.50IU/mg,酸沉淀时将pH值调节至6.3,离心上清夜中人凝血因子Ⅷ比活性为11.50 IU/mg。经过以上两步处理,可将人凝血因子Ⅷ比活性提高近2倍。结论人凝血因子Ⅷ提取工艺优化研究工艺合理、可行,能有效达到纯化提取人凝血因子Ⅷ的效果。

목적:우화혈장중인응혈인자Ⅷ제취공예조건。방법이인혈장랭침정위원료,인응혈인자Ⅷ적비활성회수솔작위평개지표,채용단인소시험,대려효적가량,려효흡부시적pH치、산침정시적pH치등진행연구,우선제취공예。결과가입랭침정중량12%적려효,병장pH치조절지7.1진행흡부,상청액중인응혈인자Ⅷ비활성가체9.50IU/mg,산침정시장pH치조절지6.3,리심상청야중인응혈인자Ⅷ비활성위11.50 IU/mg。경과이상량보처리,가장인응혈인자Ⅷ비활성제고근2배。결론인응혈인자Ⅷ제취공예우화연구공예합리、가행,능유효체도순화제취인응혈인자Ⅷ적효과。
Objective Optimal human coagulation factor Ⅷ in plasma extracts process cond-itions. Methods With human plasma cryoprecipitate as raw materials,human factor VIII activity recovery rate as the evaluation index,the single factor test was used to study the amount of alumina,the adsorption of pH value,pH value of acid precipitation,optimum extraction process. Re-sults Add cold precipitation of 12% alumina gel of 3% and the pH value is adjusted to 7.1,then adsorbed. The human factor Ⅷspecific activity ratio can reach 9.50IU/mg in the supernatant,acid precipitation when the pH value is adjusted to 6.3,human fac-torⅧspecific activity was 11.50 IU/mg in the centrifugal supernatant. Af-ter the above two steps,the human factorⅧspecific ac-tivity increased nearly 2 times. Conclusion The optimization process is reasonable,feasible,could reach purification,extra-ction of human coagulation factorⅧ.
