目的:探讨改良愈疡方对右旋葡聚糖硫酸钠( DSS)诱导的溃疡性结肠炎( UC)模型小鼠肠道菌群的影响及治疗作用。方法40只雄性C57 BL/6小鼠按完全随机法分为正常组、模型组、治疗组、对照组各10只;正常组和模型组均用0.9%氯化钠注射液灌肠,治疗组给予改良愈疡方灌肠,对照组给予柳氮磺胺吡啶灌肠,共治疗14 d。观察结肠长度和质量变化,以及对UC小鼠细菌移位的影响。结果治疗组、对照组疾病活动指数(DAI)评分与模型组比较差异均有统计学意义(P<0.01);治疗组与对照组比较差异有统计学意义(P<0.05);治疗组与正常组比较差异无统计学意义(P>0.05)。饮用DSS后各组小鼠结肠长度均有不同程度缩短,治疗组与模型组比较差异有统计学意义(P<0.01),但与正常组相比较无统计学意义(P>0.05),对照组与模型组比较差异有统计学意义( P<0.01);各组小鼠结肠质量比较,治疗组与正常组、对照组、模型组比较差异均有统计学意义(P<0.01)。结肠长度与质量之比比较,模型组明显低于正常组(P<0.01),治疗组高于模型组和对照组(P<0.01,P<0.05),正常组与治疗组比较差异无统计学意义(P>0.05)。小鼠肠系膜淋巴结细菌移位率模型组明显高于正常组、对照组(P<0.01),治疗组未发现细菌移位,与正常组比较差异无统计学意义( P>0.05)。结论中药灌肠有助于维持UC模型小鼠的肠道正常菌群,对实验性模型小鼠结肠黏膜屏障具有保护作用。

목적:탐토개량유양방대우선포취당류산납( DSS)유도적궤양성결장염( UC)모형소서장도균군적영향급치료작용。방법40지웅성C57 BL/6소서안완전수궤법분위정상조、모형조、치료조、대조조각10지;정상조화모형조균용0.9%록화납주사액관장,치료조급여개량유양방관장,대조조급여류담광알필정관장,공치료14 d。관찰결장장도화질량변화,이급대UC소서세균이위적영향。결과치료조、대조조질병활동지수(DAI)평분여모형조비교차이균유통계학의의(P<0.01);치료조여대조조비교차이유통계학의의(P<0.05);치료조여정상조비교차이무통계학의의(P>0.05)。음용DSS후각조소서결장장도균유불동정도축단,치료조여모형조비교차이유통계학의의(P<0.01),단여정상조상비교무통계학의의(P>0.05),대조조여모형조비교차이유통계학의의( P<0.01);각조소서결장질량비교,치료조여정상조、대조조、모형조비교차이균유통계학의의(P<0.01)。결장장도여질량지비비교,모형조명현저우정상조(P<0.01),치료조고우모형조화대조조(P<0.01,P<0.05),정상조여치료조비교차이무통계학의의(P>0.05)。소서장계막림파결세균이위솔모형조명현고우정상조、대조조(P<0.01),치료조미발현세균이위,여정상조비교차이무통계학의의( P>0.05)。결론중약관장유조우유지UC모형소서적장도정상균군,대실험성모형소서결장점막병장구유보호작용。
Objective To investigate the effect of modified Yuyang prescription on intestinal flora in mice model with ulcerative colitis induced by dextran sodium sulfate (DSS).Methods 40 male C57BL /6 mice were randomly divided into normal group ,model group ,treatment group and control group ,10 mice in each group; normal group and model group were given 0.9%sodium chloride injection enema ,enema of traditional Chinese medicine was used in treatment group ,the control group was given sulfasalazine enema for 14d.The changes of colon length and quality and bacterial translocation in UC mice were observed .Results There was significant difference between treatment group,control group and model group on the score of disease activity index (DAI) (P<0.01).There was significant difference between treatment group and control group on the score of DAI (P <0.05).There was no difference between treatment group and normal group on the score of DAI (P>0.05).The colon length in each group was shorten in differ degrees after the treatment of DSS .There was significant difference between treatment group and model group on the colon length (P<0.01).But there was no difference between treatment group and nor-mal group on the colon length (P>0.05).There was significant difference between control group and model group on the colon length (P<0.01).There was significant difference between treatment group ,normal group,control group and model group on the colon quality (P<0.01).The ratio of colon length and quality in model group was lower than that in normal group (P<0.01).The ratio of colon length and quality in treatment group was higher than that in model and normal group (P<0.01,P<0.05).There was no difference between treatment group and normal group on the ratio of colon length and quality (P>0.05). Mesenteric lymph node bacterial translocation rate in model group was higher than in normal group and control group (P<0.01).There was no bacterial translocation in treatment group and there was no difference between treat-ment group and normal group ( P >0.05).Conclusion Chinese medicine enema contributes to maintenance of the intestinal tract normal bacteria flora in mice model with ulcerative colitis ,and has protection on colonic mucosal barrier of experimental model mice .
