目的:研究整合素连接激酶(ILK)和血管紧张素Ⅱ(Ang-Ⅱ)在UUO大鼠模型的表达及真武汤对肾间质纤维化(RIF)的干预作用。方法采用单侧输尿管梗阻(UUO)方法复制大鼠肾间质纤维化模型,随机分为假手术组和造模组,造模组分为模型组、真武汤低剂量组、中剂量组、高剂量组和尿毒清对照组五组;分别给予相应药物灌服,术后2、3和4周分批取材,收集其尿液、血清,生化法检测24 h尿蛋白定量,ELISA法检测ILK和Ang-Ⅱ的蛋白表达。结果真武汤能够降低UUO大鼠模型血清中ILK和Ang-Ⅱ蛋白含量,具有统计学意义,真武汤中剂量组ILK和Ang-Ⅱ的含量蛋白最低。结论真武汤抑制了UUO大鼠模型血清中Ang-Ⅱ和ILK蛋白的表达,减缓了肾间质纤维化的发展和进程。

목적:연구정합소련접격매(ILK)화혈관긴장소Ⅱ(Ang-Ⅱ)재UUO대서모형적표체급진무탕대신간질섬유화(RIF)적간예작용。방법채용단측수뇨관경조(UUO)방법복제대서신간질섬유화모형,수궤분위가수술조화조모조,조모조분위모형조、진무탕저제량조、중제량조、고제량조화뇨독청대조조오조;분별급여상응약물관복,술후2、3화4주분비취재,수집기뇨액、혈청,생화법검측24 h뇨단백정량,ELISA법검측ILK화Ang-Ⅱ적단백표체。결과진무탕능구강저UUO대서모형혈청중ILK화Ang-Ⅱ단백함량,구유통계학의의,진무탕중제량조ILK화Ang-Ⅱ적함량단백최저。결론진무탕억제료UUO대서모형혈청중Ang-Ⅱ화ILK단백적표체,감완료신간질섬유화적발전화진정。
Objective To research the effect of Zhenwu decoction on the expression of integrin linked kinase and angiotensin II,l and the renal interstitial fibrosis in UUO rats.Methods The rat model of renal interstitial fibro-sis was successfully established by unilateral ureteral obstruction(UUO).The rats were divided into the control group and the model group,and the Zhenwu decoction group (low dose,middle dose,high dose)and Niao Du Qing granules control group.The corresponding drug were administrated,and the urine and serum were collected after 2,3and 4 weeks.The level of 24 hours urinary protein were tested by biochemical testing and the expression of integrin-linked kinase and angiotensionⅡwere determined by ELASA.Results Zhenwu decoction could significantly reduce the ex-pression of ILK and Ang-II in UUO rat model,and the lowest level of ILK and Ang- II were observed in the middle dose group.Conclusion Zhenwu decoction can inhibite the expression of Ang-Ⅱ and ILK in the UUO rats model, and slow the progress of renal interstitial fibrosis.
