背景:干细胞致瘤性研究是临床应用干细胞安全性所面临的实际问题。因此明确干细胞是否具有致瘤性尤为重要。裸鼠在肿瘤学、免疫学、药品及生物制品的安全性评价领域占据愈来愈重要的地位。<br> 目的:将神经干细胞、间充质干细胞移植在Balb/c免疫缺陷裸鼠皮下,检测其是否具有致瘤性。<br> 方法:将免疫缺陷Balb/c裸鼠随机分为5组,除空白组外,阳性对照组、阴性对照组、神经干细胞组和间充质干细胞组分别将肝癌细胞HepG2、人视网膜色素上皮细胞、传代至第4代的神经干细胞和间充质干细胞接种于裸鼠皮下,于注射后12周进行解剖检查,观察裸鼠移植部位肿瘤形成情况。同时进行软琼脂克隆形成实验,考察神经干细胞和间充质干细胞在体外软琼脂克隆形成情况。<br> 结果与结论:裸鼠致瘤性实验结果显示,空白组、阴性对照组、神经干细胞组、间充质干细胞组在接种12周后,未在裸鼠注射部位形成肿瘤,病理组织学检查未见异常。软琼脂克隆形成实验结果显示,神经干细胞和间充质干细胞在软琼脂阻力介质中未表现出细胞形成克隆的能力。提示短期传代后的神经干细胞、间充质干细胞不具有致瘤性。
揹景:榦細胞緻瘤性研究是臨床應用榦細胞安全性所麵臨的實際問題。因此明確榦細胞是否具有緻瘤性尤為重要。裸鼠在腫瘤學、免疫學、藥品及生物製品的安全性評價領域佔據愈來愈重要的地位。<br> 目的:將神經榦細胞、間充質榦細胞移植在Balb/c免疫缺陷裸鼠皮下,檢測其是否具有緻瘤性。<br> 方法:將免疫缺陷Balb/c裸鼠隨機分為5組,除空白組外,暘性對照組、陰性對照組、神經榦細胞組和間充質榦細胞組分彆將肝癌細胞HepG2、人視網膜色素上皮細胞、傳代至第4代的神經榦細胞和間充質榦細胞接種于裸鼠皮下,于註射後12週進行解剖檢查,觀察裸鼠移植部位腫瘤形成情況。同時進行軟瓊脂剋隆形成實驗,攷察神經榦細胞和間充質榦細胞在體外軟瓊脂剋隆形成情況。<br> 結果與結論:裸鼠緻瘤性實驗結果顯示,空白組、陰性對照組、神經榦細胞組、間充質榦細胞組在接種12週後,未在裸鼠註射部位形成腫瘤,病理組織學檢查未見異常。軟瓊脂剋隆形成實驗結果顯示,神經榦細胞和間充質榦細胞在軟瓊脂阻力介質中未錶現齣細胞形成剋隆的能力。提示短期傳代後的神經榦細胞、間充質榦細胞不具有緻瘤性。
배경:간세포치류성연구시림상응용간세포안전성소면림적실제문제。인차명학간세포시부구유치류성우위중요。라서재종류학、면역학、약품급생물제품적안전성평개영역점거유래유중요적지위。<br> 목적:장신경간세포、간충질간세포이식재Balb/c면역결함라서피하,검측기시부구유치류성。<br> 방법:장면역결함Balb/c라서수궤분위5조,제공백조외,양성대조조、음성대조조、신경간세포조화간충질간세포조분별장간암세포HepG2、인시망막색소상피세포、전대지제4대적신경간세포화간충질간세포접충우라서피하,우주사후12주진행해부검사,관찰라서이식부위종류형성정황。동시진행연경지극륭형성실험,고찰신경간세포화간충질간세포재체외연경지극륭형성정황。<br> 결과여결론:라서치류성실험결과현시,공백조、음성대조조、신경간세포조、간충질간세포조재접충12주후,미재라서주사부위형성종류,병리조직학검사미견이상。연경지극륭형성실험결과현시,신경간세포화간충질간세포재연경지조력개질중미표현출세포형성극륭적능력。제시단기전대후적신경간세포、간충질간세포불구유치류성。
BACKGROUND:Stem celltumorigenicity is a practical issue concerning stability in the clinical application of stem cells. Therefore, it is particularly important to clear whether stem cells have tumorigenic ability or not. Nude mice occupy an increasingly important position in oncology, immunology, and safety evaluation of drugs and biological products. <br> OBJECTIVE:To observe the tumorigenicity of neural stem cells and mesenchymal stem cells in Balb/c nude immunodeficient mice. <br> METHODS:Balb/c nude mice were randomly divided into control group, negative group, positive group, neural stem cellgroup and mesenchymal stem cellgroup. HepG-2 cells, RPE cells, passage 4 neural stem cells and mesenchymal stem cells were injected subcutaneously into nude mice from different groups, respectively. After 12 weeks of injection, anatomical observation was performed to detect the tumor formation in the transplantation site. Meanwhile, soft agar colony formation assay was applied to investigate neural stem celland mesenchymal stem cellclone in vitro. <br> RESULTS AND CONCLUSION:After 12 weeks of injection, the tumorigenicity study results showed that no tumor developed in the transplantation site in the control group, negative group, neural stem cellgroup and mesenchymal stem cellgroup. Histopathologic examinations also showed no abnormality in these groups. Soft agar colony formation assay showed in soft agar resistance medium, neural stem cells and mesenchymal stem cells did not exhibit clone ability. These findings indicate that neural stem cells and mesenchymal stem cells undergoing short-term passages have no tumorigenic growth.
