目的:探讨维药迪娜口服液对四氯化碳(CCl4)致急性肝损伤小鼠肝组织丙二醛(MDA)含量、谷胱甘肽过氧化物酶(GSH-Px)活力及 Ca2+转运 ATP 酶(Ca2+-ATPase)活性的影响及其保肝作用机制。方法取小鼠60只随机分成正常对照组、模型组、阳性对照药联苯双酯组(3.75 mg/kg)及迪娜口服液低、中、高剂量组(分别为4.78、9.55、19.10 g 迪娜口服液/kg 体质量,相当于人用量95.5 g/d 的5、10、20倍),每组10只。各实验药物组及阳性对照组分别按剂量灌胃给药0.2 mL/10 g 体质量,正常对照组、模型组给予等容量蒸馏水;连续给药7 d,禁食不禁水12 h,除正常对照组外,其他各组动物腹腔注射0.15%CCl4-花生油溶液(0.1 mL/10 g)。造模24 h 后取肝脏做组织匀浆,检测 MDA 含量、GSH-Px 活力与 Ca2+-ATPase 活性的变化。结果与正常对照组比较,模型组 MDA含量明显升高,GSH-Px 活力与 Ca2+-ATPase 活性均明显下降,差异有统计学意义(P <0.05);与模型组比较,阳性对照组和迪娜各剂量组 MDA 含量均降低,GSH-Px 活力及 Ca2+-ATPase 活性提高,其中迪娜中剂量组 MDA 含量降低及组织 Ca2+-ATPase 活性提高最为明显(P <0.05),迪娜高剂量组 GSH-Px 活力上升较为明显。结论迪娜口服液对 CCl4所致小鼠急性肝损伤具有明显的保护作用,其机制可能与清除自由基损伤和保护酶活性有关。

목적:탐토유약적나구복액대사록화탄(CCl4)치급성간손상소서간조직병이철(MDA)함량、곡광감태과양화물매(GSH-Px)활력급 Ca2+전운 ATP 매(Ca2+-ATPase)활성적영향급기보간작용궤제。방법취소서60지수궤분성정상대조조、모형조、양성대조약련분쌍지조(3.75 mg/kg)급적나구복액저、중、고제량조(분별위4.78、9.55、19.10 g 적나구복액/kg 체질량,상당우인용량95.5 g/d 적5、10、20배),매조10지。각실험약물조급양성대조조분별안제량관위급약0.2 mL/10 g 체질량,정상대조조、모형조급여등용량증류수;련속급약7 d,금식불금수12 h,제정상대조조외,기타각조동물복강주사0.15%CCl4-화생유용액(0.1 mL/10 g)。조모24 h 후취간장주조직균장,검측 MDA 함량、GSH-Px 활력여 Ca2+-ATPase 활성적변화。결과여정상대조조비교,모형조 MDA함량명현승고,GSH-Px 활력여 Ca2+-ATPase 활성균명현하강,차이유통계학의의(P <0.05);여모형조비교,양성대조조화적나각제량조 MDA 함량균강저,GSH-Px 활력급 Ca2+-ATPase 활성제고,기중적나중제량조 MDA 함량강저급조직 Ca2+-ATPase 활성제고최위명현(P <0.05),적나고제량조 GSH-Px 활력상승교위명현。결론적나구복액대 CCl4소치소서급성간손상구유명현적보호작용,기궤제가능여청제자유기손상화보호매활성유관。
Objective To study the protective effect of Uyghur Medicine Dina oral liquid on CCl4-induced a-cute liver injury in mice.Methods The mice were randomly divided into normal control group and CCl4-induced acute liver injury model group,bifendate treatment group (positive control,at 3.75 mg/kg)and Dina oral liquid treatment group (small dose group,at 4.78 g/kg;middle dose group,at 9.55 g/kg and high dose group,at 19.1 g/kg).The administration of bifendate and Dina oral liquid was performed once a day for 7 days.The model,bifendate treatment and Dina oral liquid treatment groups were intragastrically given 1.5% CCl4 in salad oil solution at the dosage of 1 mL/kg to induce acute liver injury,and normal control group intraperitoneal injection of the same dose of vegetable oil.At 24 hour after the administration of CCl4 ,all the mice were sacrificed to assay antioxidant activities in the blood.Meanwhile,hepatic his-topathological changes were also examined.Results The MDA level in the liver and serum of acute hepatic injury model group were significant higher than those in normal control group,while the content of the GSH-Px and Ca2+-ATPase were significantly lower than that in model control group,compared with the acute hepatic injury model group,the MDA levels in the liver in Dina oral liquid all groups were both evi-dently decreased,especially in middle dose group;while the content of GSH-Px and Ca2+-ATPase were significantly higher than that in model control group,particularly in Dina oral liquid middle dose group (Ca2+-ATPase)and in high dose group (GSH-Px).Conclusion Dina oral liquid have protective effects on acute hepatic injury induced by CCl4 in mice.
