目的观察三维适形调强放疗(IMRT)对鼠继发性骨棘球蚴的杀灭效果及周围神经的损伤,探讨IMRT对骨棘球蚴病治疗的有效性及安全性。方法选取股骨继发性棘球蚴病大鼠50只,对50只大鼠股骨棘球蚴病区给予剂量40 Gy的IMRT放疗,于放疗1 w后活杀取材,取大鼠股骨棘球蚴包囊,应用Western blot法定量检测骨棘球蚴包囊中凋亡相关蛋白Caspase-3的表达情况,取相同部位、相同位置的坐骨神经,行 HE染色、电镜观察坐骨神经神经元的变化。结果 IMRT作用1 w后,Western blot法检测到大鼠股骨棘球蚴包囊的细胞中凋亡相关基因Caspase-3大量表达,坐骨神经神经元结构正常,未受到放射线损伤。结论 IMRT对鼠继发性骨棘球蚴病有很好的治疗效果,而且有很高的安全性。

목적관찰삼유괄형조강방료(IMRT)대서계발성골극구유적살멸효과급주위신경적손상,탐토IMRT대골극구유병치료적유효성급안전성。방법선취고골계발성극구유병대서50지,대50지대서고골극구유병구급여제량40 Gy적IMRT방료,우방료1 w후활살취재,취대서고골극구유포낭,응용Western blot법정량검측골극구유포낭중조망상관단백Caspase-3적표체정황,취상동부위、상동위치적좌골신경,행 HE염색、전경관찰좌골신경신경원적변화。결과 IMRT작용1 w후,Western blot법검측도대서고골극구유포낭적세포중조망상관기인Caspase-3대량표체,좌골신경신경원결구정상,미수도방사선손상。결론 IMRT대서계발성골극구유병유흔호적치료효과,이차유흔고적안전성。
Objective To observe the possibility of the killing effect of intensity modulated radiation therapy (IMRT)to the protoscolex of bone echinococcosis in rats,and explore the killing effectiveness and safety of IMRT to the protoscolex of bone echinococcosis in rats.Methods 50 purebred bone echinococcosis rats were selected and the rats were irradiated by the models of intensity modulated radiotherapy ,the total ir-radiation doses were 40 Gy.The left and right samples of echinococcosis were taken out from the bone on first week after radiation.Extract the germinal layer cells,evaluated the expression of apoptosis-associated of Caspase-3 by Western blotting .The samples of sciatic nerve were taken out from the same locum of the echinococcosis of bone.All the samples were observed by the hematoxylin and eosin stain and electron mi-croscope.Results One week later,after treatment with intensity modulated radiation therapy,the expres-sion of Caspase-3 in echinococcosis of bone was up-regulated.The neurons in the peripheral nerve of IMRT group were mainly irreversible inj ury and mainly apoptosis.Conclusion It is evident that the bone echino-coccosis cells in rats can be damaged by IMRT,and has high security.
