目的:观察电针治疗对慢性应激抑郁模型大鼠行为学、血清褪黑素(melatonin,MT)以及松果体芳基烷化胺N-乙酰基转移酶(arylalkylamine N-acetyltransferase,AANAT)mRNA、羟基吲哚氧位甲基转移酶(hydroxyindole-O-methyl-transferase,HIOMT)mRNA表达的影响,探讨电针对慢性应激抑郁模型大鼠血清MT含量调节的作用机制,从而进一步充实电针抗抑郁的作用机理。方法将大鼠随机分为3组,空白组、模型组、电针组,每组10只,共30只,通过慢性温和不可预知性应激刺激结合孤养的方法复制慢性应激抑郁模型,电针组给予强度为2 Hz、1 mA的电针治疗,每次20分钟,每天1次,共电针21天。采用开野试验观察大鼠的行为学变化;采用酶联免疫吸附法对大鼠血清中MT的含量进行检测,用Real-time PCR检测各组大鼠松果体AANAT mRNA、HIOMT mRNA的表达情况。结果在大鼠开野试验中,与空白组比较,模型组大鼠水平运动、垂直运动的次数均显著减少(P=0.000<0.01,P=0.007<0.01),而电针可逆转此变化,与模型组相比,电针组大鼠的水平运动次数和垂直运动次数均显著增多(P=0.000<0.01,P=0.001<0.01);模型组大鼠血清中MT含量较空白组明显降低(P=0.000<0.01),与模型组大鼠比较,电针可提高血清中MT含量(P=0.000<0.01)。与空白组相比,模型组 AANAT mRNA、HIOMT mRNA的表达均显著降低(P =0.000<0.01,P=0.026<0.05),而电针组的表达较模型组均显著升高(P =0.000<0.01,P =0.000<0.01)。结论电针可以提高慢性应激抑郁模型大鼠血清中 MT含量,并且可以提高AANAT mRNA、HIOMT mRNA的表达。其中,电针提高慢性应激抑郁模型大鼠AANAT mRNA、HI-OMT mRNA的表达可能是提高大鼠血清中MT含量的机制之一。

목적:관찰전침치료대만성응격억욱모형대서행위학、혈청퇴흑소(melatonin,MT)이급송과체방기완화알N-을선기전이매(arylalkylamine N-acetyltransferase,AANAT)mRNA、간기신타양위갑기전이매(hydroxyindole-O-methyl-transferase,HIOMT)mRNA표체적영향,탐토전침대만성응격억욱모형대서혈청MT함량조절적작용궤제,종이진일보충실전침항억욱적작용궤리。방법장대서수궤분위3조,공백조、모형조、전침조,매조10지,공30지,통과만성온화불가예지성응격자격결합고양적방법복제만성응격억욱모형,전침조급여강도위2 Hz、1 mA적전침치료,매차20분종,매천1차,공전침21천。채용개야시험관찰대서적행위학변화;채용매련면역흡부법대대서혈청중MT적함량진행검측,용Real-time PCR검측각조대서송과체AANAT mRNA、HIOMT mRNA적표체정황。결과재대서개야시험중,여공백조비교,모형조대서수평운동、수직운동적차수균현저감소(P=0.000<0.01,P=0.007<0.01),이전침가역전차변화,여모형조상비,전침조대서적수평운동차수화수직운동차수균현저증다(P=0.000<0.01,P=0.001<0.01);모형조대서혈청중MT함량교공백조명현강저(P=0.000<0.01),여모형조대서비교,전침가제고혈청중MT함량(P=0.000<0.01)。여공백조상비,모형조 AANAT mRNA、HIOMT mRNA적표체균현저강저(P =0.000<0.01,P=0.026<0.05),이전침조적표체교모형조균현저승고(P =0.000<0.01,P =0.000<0.01)。결론전침가이제고만성응격억욱모형대서혈청중 MT함량,병차가이제고AANAT mRNA、HIOMT mRNA적표체。기중,전침제고만성응격억욱모형대서AANAT mRNA、HI-OMT mRNA적표체가능시제고대서혈청중MT함량적궤제지일。
Objective Observe the influence of electro-acupuncture of the locomotor activity ser-um MT and AANAT mRNA,HIOMT mRNA of chronic unpredictable mild stress depression model rats. Discuss the biological mechanisms of the effect of electro-acupuncture on the serum MT chronically and the biological mechanisms of electro-acupuncture treating depression. Methods 30 rats are randomly divided into 3 groups:blank group,model group,EA(electro-acupuncture)group,10 in each group. Use the chronic unpredictable mild stress combined with solitarily feeding method to make the depression model. Treat the EA group with stimulation by electro-acupuncture;the stimulus intensity of current strength is 1 mA,2 hz and 20 minutes,once every day for a period of 21 days. Observe the locomotor activity of rats in every group by open-field experiment. Observe the content of the serum MT using ELISA. Detect AANAT mRNA and HIOMT mRNA in the pineal body of each group by real-time PCR. Result The times of cross and rear movement of the model group are significantly lower than blank group(P=0.000<0.01、P=0.007<0.01),while electro-acupuncture can reverse it,compared with the model group,the times of cross and rear movement of the EA (electro-acupuncture)group are significantly increased(P=0.000<0.01,P=0.001 <0.01). Compared with the blank group,the content of serum MT significantly de-creased in model group (P=0.000<0.01),compared with the model group,electro-acupuncture can in-crease the content of serum MT (P=0.000<0.01). Compared with the blank group,the expression of HIOMT mRNA and AANAT mRNA of model group,were significantly lower (P=0.000<0.01、P=0.026<0.05),electro-acupuncture can significantly increase the expression of HIOMT mRNA and AANAT mR-NA (P=0.000<0.01,P=0.000<0.01). Conclusion Electro-acupuncture can improve the contents of serum MT in chronic unpredictable mild stress depression model rats,and can improve the expression of AANAT mRNA and HIOMT mRNA. The improvement of the expression of AANAT mRNA and HIOMT mRNA may be one of the mechanisms to improving the content of serum MT of chronic stress depression model rats.
