目的探讨E-cadherin的表达和AKT的磷酸化水平与宫颈病变程度的相关性。方法收集20例正常宫颈组织、30例宫颈上皮内瘤变( CIN )组织、50例宫颈鳞癌组织,采用荧光实时定量PCR和免疫组化方法分别检测组织中E-cadherin、β-catenin、p-AKT 和 Loricrin 的 mRNA 及蛋白表达情况,分析E-cadherin表达和AKT磷酸化水平的相关性,及其与宫颈病变程度和宫颈鳞癌分化程度的相关性。结果免疫组化结果显示,随着宫颈病变的进展,E-cadherin、Loric-rin的表达逐渐降低,甚至缺失( P<0.05);在正常宫颈上皮β-catenin表达于胞膜,随着宫颈病变的进展,逐渐由胞膜向胞质和胞核转移,且表达阳性率逐渐降低( P<0.05);上述指标定量PCR结果与免疫组化结果基本一致。 p-AKT的表达则随病变进展而逐渐升高(P<0.05)。相关分析显示,E-cadherin的表达与宫颈病变的程度及鳞癌的分化程度呈负相关(rs =-0.688,-0.438;P<0.01),而与p-AKT的表达则呈正相关( rs =0.462,0.692;P <0.01);E-cadherin 与p-AKT的表达呈负相关(rs =-0.828, P<0.01)。结论 E-cadherin的表达缺失与AKT磷酸化水平的增高与宫颈病变的发生发展密切相关;二者联合可用于监测宫颈病变的进展,指导临床治疗方案。

목적탐토E-cadherin적표체화AKT적린산화수평여궁경병변정도적상관성。방법수집20례정상궁경조직、30례궁경상피내류변( CIN )조직、50례궁경린암조직,채용형광실시정량PCR화면역조화방법분별검측조직중E-cadherin、β-catenin、p-AKT 화 Loricrin 적 mRNA 급단백표체정황,분석E-cadherin표체화AKT린산화수평적상관성,급기여궁경병변정도화궁경린암분화정도적상관성。결과면역조화결과현시,수착궁경병변적진전,E-cadherin、Loric-rin적표체축점강저,심지결실( P<0.05);재정상궁경상피β-catenin표체우포막,수착궁경병변적진전,축점유포막향포질화포핵전이,차표체양성솔축점강저( P<0.05);상술지표정량PCR결과여면역조화결과기본일치。 p-AKT적표체칙수병변진전이축점승고(P<0.05)。상관분석현시,E-cadherin적표체여궁경병변적정도급린암적분화정도정부상관(rs =-0.688,-0.438;P<0.01),이여p-AKT적표체칙정정상관( rs =0.462,0.692;P <0.01);E-cadherin 여p-AKT적표체정부상관(rs =-0.828, P<0.01)。결론 E-cadherin적표체결실여AKT린산화수평적증고여궁경병변적발생발전밀절상관;이자연합가용우감측궁경병변적진전,지도림상치료방안。
Objective To investigate the correlation between the expression of E-cadherin and the level of phospho-rylation of AKT and the degree of cervical lesions. Methods 20 cases of normal cervical tissue, 30 cases of CIN tissues, and 50 cases of cervical squamous cell carcinoma were collected;real-time PCR and immunohistochemistry were used to detect mRNA and protein expression of E-cadherin,β-catenin, p-AKT and Loricrin in tissue, correla-tion between E-cadherin expression and AKT phosphorylation levels,and the correlation between two markers above and degree of cervical lesions and the differentiation of cervical cancer were analysed. Results Immunohistochemi-cal results showed that with the progress of cervical lesions, expression of E-cadherin and Loricrin would decrease, or absent ( P<0. 05 ); β-catenin expresses in the membrane in normal epithelial, gradually transferring from the membrane to the cytoplasm and nucleus with cervical lesions progressing, and the expression was totally decreased ( P<0. 05 ); quantitative PCR results were consistent with the above-mentioned immunohistochemical results of protein. Conversly, the expression of p-AKT gradually increased with the progress of the disease (P<0. 05). The analyses showed that the expression of E-cadherin and stage of cervical lesions and squamous differentiation were negatively correlated (rs = -0. 688,-0. 438; P<0. 01) while the expression of p-AKT had positive correlation with them (rs=0. 462, 0. 692; P <0. 01). E-cadherin and p-AKT expression were negatively correlated (rs =-0. 828,P<0. 01). Conclusion Loss of E-cadherin expression and increase of AKT phosphorylation are closely related with the development of cervical lesions;the combination of them can be used to monitor the progress of cer-vical lesions and guide clinical treatment programs.
