真核生物新生mRNA前体(pre-mRNA)经过5'戴帽、剪接和3'加尾等过程加工为成熟的mRNA。一个pre-mRNA通过选择不同的剪接位点组合产生不同的mRNA剪接变异体(Splice variant),从而可由一个基因产生若干具有独特结构和功能的蛋白异构体(isoforms)。研究显示pre-mRNA的选择性剪接是控制基因表达和产生蛋白质多样性的重要机制,与许多疾病的发生密切相关。我们将对基因转录剪接体在疾病的发生与诊断治疗中的作用及最新进展进行探讨。

The mature mRNA came into being with the 5' in cap and splicing and 3' end processing of the pre-mRNA of new precursors of eukaryotes. A pre-mRNA can make different mRNA splicing variants by choosing different combinations of splicing sites, which makes a gene able to produce several protein isoforms with unique structure and function. The study shows that the selective splicing of the pre-mRNA is an important mechanism for the control of gene expression and protein diversity, which is closely related to the occurrence of many diseases. We will discuss on the significant role of gene transcription splicing in the diagnosis and treatment of the disease and its latest progress.
