目的 观察人脐带间充质干细胞(hUCMSCs)联合布洛芬移植治疗大鼠脊髓损伤的效果,探讨布洛芬对hUCMSCs存活和分化的影响.方法 用Impactor ModelⅡ打击器制作80例大鼠脊髓胸10(T10)损伤模型,随机分为DMEM实验对照组(A组)、布洛芬注射组(B组)、单纯移植hUCMSCs组(C组)、联合治疗组(D组),对损伤后2个月内的实验大鼠进行行为学评分(BBB评分),评估后肢运动功能恢复,结合人细胞核抗体(MAB1281)和神经丝蛋白(NF-200)免疫组织化学染色分析细胞存活和分化.结果 行为学评分D组(14.90±2.45)分,明显高于B组(10.56±2.32)分、C组(11.90±1.89)分,差异有统计学意义(P<0.05).术后8周观察到D组的hUCMSCs存活数量多于其他治疗组;NF-200染色面积D组(0.28±0.13)高于B组(0.11±0.04)、C组(0.18 ±0.06),差异有统计学意义(P<0.05).结论 布洛芬可以促进hUCMSCs在损伤部位存活并使其易于向神经样细胞分化,两者联合移植可以明显促进损伤后轴突再生和脊髓功能恢复.

Objective To investigate the effects of ibuprofen combined with transplantation of human umbilical cord mesenchymal stem cells (hUCMSCs) in rats with spinal cord injury,and explore the influence of ibuprofen on the survival and differentiation of hUCMSCs.Methods Eighty SCI models were made by Impactor Model Ⅱ at T10,and were randomized into 4 groups:DMEM control (group A),Ibuprofen group (group B),hUCMSCs group (group C),co-transplantation (group D).The basso beattie bresnahan (BBB) score was practised to evaluate functional recovery of rat's hind limbs,and immunohistochemistry was used to observe the survival and differentiation of hUCMSCs.Results Functional recovery was more advanced in group D (14.90 ±2.45) points than in group B (10.56 ±2.32) points or group C (11.90 ± 1.89) points according to The BBB scores (P < 0.05).Immunohistochemical staining showed the number of surviving hUCMSCs was more in group than other group,and the NF-200 positive area in cotransplantation at 8th week after transplantation was 0.28 ± 0.13 in group D,greater than in group B (0.11 ± 0.04) or group C (0.18 ± 0.06) (P < 0.05).Conclusion Ibuprofen can promote the survival and differentiation of the transplanted hUCMSCs.Co-transplantation of hUCMSCs and ibuprofen may significantly promote axonal regeneration and functional recovery after SCI.
