采用乙酸和酸性蛋白酶脱除虾头虾壳中的矿物质和蛋白质,研究提取甲壳素的工艺条件。通过比较发现SUKA酸性蛋白酶脱除蛋白质的效果最好,并且发现脱除蛋白质有利于脱除矿物质。在此基础上进一步研究温度、 SUKA酶的添加量、 pH值、料液比、反应时间对提取甲壳素的影响,并采用响应面法优化工艺条件。结果表明,提取甲壳素的最佳工艺条件为SUKA酶的添加量0.2%,温度46℃,反应pH值4.0,料液比1∶20,反应时间48 h,在此条件下得到的甲壳素中蛋白质含量6.52%,灰分含量1.91%。

In this paper shrimp waste is demineralized and deprotenized with acetic acid and acid protease to extract chitin. The technological parameters are investigated. It is found that SUKA acid protease is most effective on deproteinization among all proteases tested, and the removal of protein is helpful for the removal of minerals. Furthermore, the effects of enzymic dosage, reaction temperature, pH, solid-to-liquid ratio and reaction time on chitin extraction are studied, and the response surface method is employed to optimize the reaction condition. The results show that the optimal conditions are as follows: enzyme dosage 0.2%, temperature 46℃, initial pH 4.0, solid-to-liquid ratio 1∶20 and reaction time 48 h. Under such conditions, the obtained chitin contains 6.52%protein and 1.91%minerals.
