目的:探讨金雀异黄素(Gen)对氧化损伤的人晶状体上皮细胞(HLEC)凋亡有无抑制作用。方法:本研究在终浓度3×10-4mol/L H2O2培养液中复制HLEC凋亡模型,并以雌二醇(E2)作为阳性对照,采用流式细胞仪(FCM)检测HLEC凋亡率和线粒体膜电位(ΔΨM)的变化。结果:FCM结果显示:H2 O2组HLEC凋亡率显著高于空白对照组;E2和Gen组凋亡率均低于H2O2组(P<0.01)。 H2O2组线粒体膜电位比对照组显著下降;E2组和Gen组的线粒体膜电位均比H2O2组升高(P<0.01)。结论:Gen可减轻实验性氧化损伤的HLEC凋亡程度。

목적:탐토금작이황소(Gen)대양화손상적인정상체상피세포(HLEC)조망유무억제작용。방법:본연구재종농도3×10-4mol/L H2O2배양액중복제HLEC조망모형,병이자이순(E2)작위양성대조,채용류식세포의(FCM)검측HLEC조망솔화선립체막전위(ΔΨM)적변화。결과:FCM결과현시:H2 O2조HLEC조망솔현저고우공백대조조;E2화Gen조조망솔균저우H2O2조(P<0.01)。 H2O2조선립체막전위비대조조현저하강;E2조화Gen조적선립체막전위균비H2O2조승고(P<0.01)。결론:Gen가감경실험성양화손상적HLEC조망정도。
Objective:To investigate if Genistein(Gen) inhibits the apoptosis of human lens epithelial cell (HLEC) resulted from the ex-perimental oxidative injury .Methods:HLECs were incubated with 3 ×10 -4 mol/L H2 O2 .And make estradiol ( E2 ) as a positive con-trol.HLECs apoptosis and apoptosis rate were measured by FCM .Results:Apoptosis rate in H2 O2 group was significandy higher than that in control group , E2 and Gen group .The mitochondria transmembrane potential in H 2 O2 group reduced obviously , comparing to control group, E2 and Gen group.Conclusion:Gen may decrease the apoptosis of HLEC resulted from the experimental oxidative inju-ry, providing scientific evidence for pursue effective medicine to prevent and treat cataract .
