利用高效液相色谱法(RP-HPLC-DAD),建立同时测定陈皮中3,5,6,7,8,3′,4′-七甲氧基黄酮、8-羟基-3,5,6,7,3′,4′-六甲氧基黄酮和5-羟基-3,6,7,8,3′,4′-六甲氧基黄酮质量分数的定量分析法,并采用 Cosmosil C18-Ar-Ⅱ(4.6 mm ×250 mm,5μm )色谱柱, V (乙腈)∶V (ρ(醋酸)=0.2%)为流动相进行梯度洗脱,流速为1.0 mL/min,检测波长为258 nm,柱温25℃.结果表明:3,5,6,7,8,3′,4′-七甲氧基黄酮的线性范围为0.01~10.0μg (r 2=0.9992),平均回收率为99.7%,相对标准偏差(RSD)为0.50%;8-羟基-3,5,6,7,3′,4′-六甲氧基黄酮的线性范围为0.05~10.0μg (r 2=0.9994),平均回收率为101.8%, RSD为2.0%;5-羟基-3,6,7,8,3′,4′-六甲氧基黄酮的线性范围为0.025~5.0μg (r 2=0.9998),平均回收率为100.2%,RSD为1.7%.

An RP-HPLC-DAD method was established for the determination of 3,5,6,7,8,3′,4′-heptamethoxyflavone,8-hydroxy-3,5,6,7,3′,4′-hexamethoxyflavone and 5-hydroxy-3,6,7,8,3′,4′-hexamethoxyflavone in citri reticulatae pericarpium.A cosmosil C18-Ar-Ⅱ(4.6 mm × 250 mm, 5μm)column was used.The mobile phase consisted of V (acetonitrile)∶V (ρ(acetic acid)=0.2%) was in the process of gradient elution.Flow rate was 1.0 mL/min,the wavelength of DAD detector was set at 258 nm and the column temperature was 25 ℃.The linear range of 3,5,6,7,8,3′,4′-heptamethoxyflavone was 0.01-10.0 μg (r 2=0.999 2 ),and the mean recovery rate was 99.7%(RSD = 0.50%). The linear range of 8-hydroxy-3,5,6,7,3′,4′-hexamethoxyflavone was 0.05-10.0μg(r 2=0.999 4),and the mean recovery rate was 101.8% (RSD=2.0%).The linear range of 5-hydroxy-3,6,7,8,3′,4′-hexamethoxyflavone was 0.025-5.0μg(r 2=0.999 8),and the mean recovery rate was 100.2% (RSD=1.7%).
