目的:探讨Smac、Survivin和PTEN在子痫前期患者胎盘组织和外周血中的表达及其相关性。方法:采用RT-PCR、免疫组化SP法测定90例胎盘组织(其中正常孕妇组、轻度和重度子痫前期组各30例)中Smac、Sur-vivin和PTEN mRNA和蛋白的表达,ELISA法测定外周血中Smac、Survivin和PTEN蛋白的水平。结果:①Smac、Sur-vivin主要定位于绒毛细胞滋养细胞胞浆中,PTEN主要定位于绒毛滋养细胞胞核中。②轻、重度子痫前期组胎盘及外周血中Smac mRNA和蛋白表达水平均高于对照组,且重度组高于轻度组,差异均有统计学意义( F=19.874和112.774,P<0.001)。③轻、重度组子痫前期组胎盘及外周血中Survivin、PTEN mRNA和蛋白表达水平均低于对照组,且重度组低于轻度组,差异均有统计学意义(F=21.306、23.485和152.751、630.848,P<0.001)。④子痫前期组胎盘和外周血中Smac与Survivin及PTEN蛋白表达呈负相关(P<0.05)。⑤子痫前期组胎盘和外周血中Smac蛋白与新生儿体重及胎盘质量呈负相关,Survivin和PTEN蛋白与新生儿体重及胎盘质量呈正相关(P<0.05)。结论:Smac高表达、Survivin和PTEN低表达在子痫前期的发生、发展中起重要作用,与新生儿预后有关。

목적:탐토Smac、Survivin화PTEN재자간전기환자태반조직화외주혈중적표체급기상관성。방법:채용RT-PCR、면역조화SP법측정90례태반조직(기중정상잉부조、경도화중도자간전기조각30례)중Smac、Sur-vivin화PTEN mRNA화단백적표체,ELISA법측정외주혈중Smac、Survivin화PTEN단백적수평。결과:①Smac、Sur-vivin주요정위우융모세포자양세포포장중,PTEN주요정위우융모자양세포포핵중。②경、중도자간전기조태반급외주혈중Smac mRNA화단백표체수평균고우대조조,차중도조고우경도조,차이균유통계학의의( F=19.874화112.774,P<0.001)。③경、중도조자간전기조태반급외주혈중Survivin、PTEN mRNA화단백표체수평균저우대조조,차중도조저우경도조,차이균유통계학의의(F=21.306、23.485화152.751、630.848,P<0.001)。④자간전기조태반화외주혈중Smac여Survivin급PTEN단백표체정부상관(P<0.05)。⑤자간전기조태반화외주혈중Smac단백여신생인체중급태반질량정부상관,Survivin화PTEN단백여신생인체중급태반질량정정상관(P<0.05)。결론:Smac고표체、Survivin화PTEN저표체재자간전기적발생、발전중기중요작용,여신생인예후유관。
Aim:To study the expressions and correlations of the mitochondrial protein Smac , antiapoptotic gene Sur-vivin and tumor suppressor PTEN in placentae and maternal blood from patients with preeclampsia .Methods:A total of 90 placental samples (normal pregnancies=30, mild preeclampsia=30, severe preeclampsia =30) were collected.The ex-pressions of Smac , Survivin and PTEN were measured by reverse transcriptase PCR and immunohistochemical SP method and ELISA.Results:Smac and Survivin were mainly expressed in trophoblast cytoplasm ,and PTEN was mainly expressed in trophoblast nucleus .The expressions of Smac mRNA and protein in placentae and maternal peripheral blood from pa-tients with mild and severe preeclampsia were significantly higher than those from control group ,moreover ,which was higher in severe preeclampsia group(F=19.874,112.774,P<0.001).The expressions of Survivin,PTEN mRNA and protein in placentae and maternal peripheral blood from patients with mild and severe preeclampsia were significantly lower than those in control group, moreover,which was lower in severe preeclampsia group (F=21.306,23.485 and 152.751,630.848,P<0.001).The expressions of Smac and Survivin ,Smac and PTEN protein were negatively correlated in preeclampsia group (P<0.05).The expression of Smac protein was negatively correlated with the weight of the newborn and the placenta , while Survivin and PTEN proteins were positively correlated with the weight of the newborn and the placenta (P<0.05). Conclusion:The abnormal expressions of Smac , Survivin and PTEN may contribute to the occurrence of preeclampsia and are related to prognosis of the birth .
